摘要
目的探讨重度子痫前期患者妊娠晚期胎盘组织中Nrf2和NQO1的表达情况及其与重度子痫前期发病的关系。方法选择重度子痫前期和正常妊娠晚期行剖宫产术的孕产妇各30例,应用Real-Time PCR法和免疫组化法分析两组孕产妇妊娠晚期胎盘组织中Nrf2和NQO1的表达情况。结果与正常组相比,重度子痫前期组胎盘组织中Nrf2和NQO1 mRNA的表达水平均降低,差异有统计学意义(P<0.05)。Nrf2主要在滋养层细胞的细胞质中表达,细胞核少见表达;NQO1蛋白在滋养层细胞的细胞质中表达。Nrf2和NQO1在重度子痫前期组胎盘组织滋养层细胞中的阳性表达率分别为46.7%和43.3%,低于正常组的76.7%和70.0%,差异有统计学意义(P<0.05)。结论胎盘组织中Nrf2和NQO1的表达水平下降可能参与了子痫前期的发病过程。
Objective To explore the expressions of Nrf2 and NQO1 in placental tissues of patients with severe preeclampsia and the relationships with onset of preeclampsia. Methods Thirty pregnant women with preeclampsia( preeclampsia group) and normal late pregnant women( control group) were selected,RT- PCR and immunohistochemical method were used to analyze the expressions of Nrf2 and NQO1 in placental tissues of the two groups. Results The expression levels of Nrf2 and NQO1 mRNA in preeclampsia group were statistically significantly lower than those in control group( P〈0.05). Nrf2 mainly expressed in cytoplasm of trophoblastic cells,while it rarely expressed in nuclei; NQO1 protein mainly expressed in cytoplasm of trophoblastic cells. The positive expression rates of Nrf2 and NQO1 in preeclampsia group were 46. 7% and 43. 3%,respectively,which were statistically significantly lower than those in control group( 76. 7% and70. 0%)( P〈0.05). Conclusion The decrease of expression levels of Nrf2 and NQO1 might be involved in the process of pathogenesis of preeclampsia.
