摘要
目的探讨子痫前期患者胎盘组织及血清中信号转导和转录激活因子4(STAT4)以及信号转导和转录激活因子6(STAT6)的表达及意义。方法 1采用Western Blot法及酶连免疫吸附试验(ELASIA)法检测30例正常晚期妊娠孕妇(对照组)和50例子痫前期患者(子痫前期组,其中轻度22例、重度28例)胎盘组织和血清中STAT4、STAT6的蛋白表达。结果 STAT4在正常妊娠组、子痫前期轻度组与重度组胎盘和血清组织中的含量随着病情的加重呈增加趋势,轻、重度子痫前期组胎盘和血清组织中STAT4的表达均高于对照组(P<0.01)。STAT6在正常妊娠组、子痫前期轻度组与重度组的胎盘及血清含量随着病情的加重呈减低的趋势,轻、重度子痫前期组胎盘和血清组织中STAT6的表达均低于对照组(P<0.01)、其中重度组STAT6表达低于轻度组(P<0.05)。子痫前期组和正常组胎盘和血清组织中STAT4与STAT6的表达水平均呈负相关。结论 STAT4、STAT6共同参与子痫前期的病理生理改变,并与病情严重症程度有一定关系。子痫前期胎盘、血清中STAT4的高表达和STAT6的低表达可能诱导T辅助淋巴细胞1/2失衡,进而导致子痫前期的发病。
Objective To explore the expressions of STAT4 and STAT6 in placenta and serum of patients with preeclampsia and the significance. Methods Western Blot and ELASA were used to detect the expressions of STAT4 and STAT6 proteins in placenta and serum of normal late pregnant women( control group) and 50 patients with preeclampsia( preeclampsia group,including 22 patients with mild preeclampsia and 28 patients with severe preeclampsia). Results The contents of STAT4 in placenta and serum of control group,mild preeclampsia group,and severe preeclampsia group showed an increasing trend with aggravation of the disease,the expressions of STAT4 in placenta and serum of mild preeclampsia group and severe preeclampsia group were statistically significantly higher than those in control group( P〈0.01). The contents of STAT6 in placenta and serum of control group,mild preeclampsia group,and severe preeclampsia group showed a decreasing trend with aggravation of the disease,the expressions of STAT6 in placenta and serum of mild preeclampsia group and severe preeclampsia group were statistically significantly lower than those in control group( P〈0.01),the expressions of STAT6 in placenta and serum of severe preeclampsia group were statistically significantly lower than those in mild preeclampsia group( P〈0.05). In placenta and serum of preeclampsia group and control group,STAT4 expression level was negatively correlated with STAT6 expression level. Conclusion STAT4 and STAT6 are involved in pathogenic process of preeclampsia,and they are correlated with severity of the disease to a certain extent. The high expression of STAT4 and low expression of STAT6 in placenta and serum may cause preeclampsia by inducing Th1 / Th2 imbalance.
