摘要
目的筛选出与子宫脱垂相关的长链非编码RNA(long non-coding RNA,lncRNA),为探讨lncRNA参与子宫脱垂的发生与发展提供实验依据。方法选取2015年1~6月在武汉市人民医院进行手术的6例子宫脱垂组织和6例未脱垂的正常子宫组织,应用高通量lncRNA芯片技术检测两组子宫组织中lncRNA表达谱的变异。结果分析两组lncRNA表达谱的变化,发现与正常子宫组织相比,子宫脱垂组织中的lncRNA和mRNA表达量均表现出上调和下调。上调mRNA(Fold〉2)参与了多细胞生物过程、信号传导和细胞交流等生物过程,下调mRNA(Fold〉2)参与了细胞器形成、细胞周期和细胞分裂等生物过程。通过GO聚类分析着重分析了变化差异性最明显的10个lncRNA,寻找出了与其变化密切相关的mRNA。通过荧光定量聚合酶链反应对4个变化最明显的lncRNA(BC008359、RP11-521B24.5、BRE-AS1和RP11-229P13.15)进一步验证,与芯片检测结果一致。结论子宫脱垂组织中lncRNA的变化情况说明,4个变化最明显的lncRNA(BC008359、RP11-521B24.5、BRE-AS1和RP11-229P13.15)在子宫脱垂组织中发挥重要作用,为探讨lncRNA参与子宫脱垂的发生与发展提供实验依据和理论基础。
Objective To screen out the long non-coding RNA( lncRNAs) related with uterine prolapse to provide experimental evidences for occurrence and development of uterine prolapse related with lncRNAs. Methods Selected 6 prolapsed uterine tissues and 6 normal uterine tissues in People's Hospital of Wuhan from January 2015 to June 2015,and applied high-through put microarray lncRNA to test the variation of lncRNAs expression profiles. Results By analyzing the changes of lncRNA expression profiles in two groups,we found that compared with normal uterine tissues,lncRNAs and mRNA expression in uterine prolapse tissues showed upregulated and down-regulated. Up-regulated mRNAs involved in the process of multi- cellular organisms,signal transduction and cell exchanges. Down- regulated mRNAs involved in the formation of organelles,cell cycle,cell division and other biological processes. The10 lncRNAs analyzed by GO showed the most obvious difference and being closely related to its mRNA. The results of fluorogenic quantitative PCR were consistent with the microarray,which suggested that the four lncRNAs( BC008359,RP11- 521B24. 5,BRE-AS1,and RP11- 229P13. 15) played important roles in uterine prolapse. Conclusion The changes of lncRNAs in the uterine prolapse showed 4 lncRNAS( BC008359、RP11- 521B24. 5、BRE- AS1 and RP11- 229P13. 15) which change most obviously play important roles in prolapsed uterine tissues,and it provides experimental and theoretical basis for investigating the occurrence and development of uterine prolapse related with lncRNAs.
