高岭土竹纤维止血纱布的制备与评价 被引量:6

Preparation and Evaluation of Kaolin Bamboo Fiber Hemostatic Gauze

摘要 目的为满足新型战场止血需要,本研究研发了高岭土竹纤维止血纱布,观察其止血功能,并对其主要成分高岭土进行生物安全性评价。方法制备4种不同浓度的高岭土竹纤维止血纱布(0%,0.1%,0.5%,1%高岭土),建立兔急性出血动物模型;测定10 min各组止血量;流式细胞术(flow cytometry,FCM)检测各组外周血中血小板活化水平;HE染色观察受压组织形态学变化及纱布颗粒脱落物数量;以人血管内皮细胞(human vascular endothelial cells,HUVEC)细胞株为研究模型,观察高岭土的细胞毒性。结果与单纯竹纤维止血纱布组比较,0.5%,1%高岭土组10 min出血量显著减少;FCM检测发现,高岭土纱布能够更好的激活外周血中血小板的活化功能,其活化程度与高岭土呈现浓度依赖性;病理切片显示,各组纱布按压止血后,创面未发现纱布颗粒脱落物,但1%高岭土组出现了部分炎性细胞浸润。体外实验证实,高岭土具有良好的生物相容性,HUVEC均能在高岭土表面生长,并不会诱导细胞凋亡。结论高岭土竹纤维止血纱布具有显著的止血效果,且高岭土生物安全性好,其中含0.5%高岭土的竹纤维止血纱布为最理想的纱布。 Objective To meet the needs of the new battlefield hemostatic,developed kaolin bamboo fiber hemostatic gauze,observed the hemostatic function and evaluated the biological safety of kaolin which was the main ingredients of gauze.Methods Producted 4 kinds of different concentration of kaolin bamboo fiber hemostatic gauze(0%,0.1%,0.5%,1% kaolin);established rabbit animals model of acute hemorrhage;detected amount of blood within 10 min;the level of peripheral blood platelet activation detected by flow cytometry(FCM);compression histomorphology changes and gauze shed particles number were observed by HE staining;human vascular endothelial cells(HUVEC)as the research model to observe the cytotoxicity of kaolin.Results Compared with pure bamboo fiber hemostatic gauze group,the bleeding within 10 min of 0.5%,1% kaolin group were significantly reduced;FCM detected that kaolin gauze could better activated the activation function of peripheral blood platelet and the activation degree with concentration-dependence;pathological biopsy showed that gauze shed particles were not found in the wound after hemostatic gauze compressions in each group,but found some inflammatory cell infiltration in 1% kaolin group.In vitro studies confirmed that the kaolin had a good biocompatibility,HUVEC could grow on the surface of kaolin and not induced apoptosis.Conclusion Kaolin bamboo fiber hemostatic gauze had obvious hemostatic effect and kaolin had a good biological safety,0.5% kaolin bamboo fiber hemostatic gauze was the most ideal gauze.
