摘要
目的:通过建立小鼠游泳和大鼠跑台2个训练实验模型,研究紫草提取物对骨骼肌自由基代谢的影响以及抗疲劳作用;并测定紫草提取物中的有效成分。方法:选取雄性小鼠和大鼠各24只,将小鼠随机分为游泳训练组(a组)、游泳力竭组(b组)和游泳加药组(c组),每组各8只;将大鼠随机分为跑台训练组(A组)、跑台力竭组(B组)、跑台加药组(C组),每组各8只。小鼠和大鼠按照不同的训练模型分别进行游泳和跑台训练,在训练方案的最后1天,a、A组动物不进行训练,在安静状态下处死制备待测样本,并通过试剂盒测定骨骼肌超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、巯基(-SH)、丙二醛(MDA)指标;b、B、c、C组动物进行一次力竭运动,力竭后即刻处死并进行样本处理和上述指标测定。结果:训练对照组(b、B组)动物在游泳和跑台力竭运动后,骨骼肌组织抗氧化酶活性显著下降(P<0.05),自由基代谢产物MDA含量显著升高(P<0.05);补充紫草提取物组(c、C组)动物在力竭运动后,与训练对照组动物(b、B)相比,骨骼肌组织抗氧化酶活性显著升高(P<0.05),自由基代谢产物MDA含量显著降低(P<0.05),运动至力竭的时间明显延长(P<0.05)。结论:外源性补充紫草提取物可以改善骨骼肌组织自由基清除和代谢水平,延缓运动疲劳的产生。
Objective:To explore the influence of gromwell extract on the metabolism of free radicals in skeletal muscle and its anti-fatigue effect by establishing two training models of mouse swimming and rat treadmill.Methods:Totally 24 male mice and 24 male rats were selected.Mice were divided into 3 groups—swimming training group(group a),exhausted swimming group(group b),swimming and medicine group(group c),and rats were divided into 3 groups—treadmill training group(group A),exhausted treadmill group(group B),treadmill training and medicine group(group C),with 8 mice or rats in each group.Mice and rats were trained by different training schemes.At the last day of training,group a and group A were not trained as usuall and were sacrificed at resting state to prepare test samples,and the determination of indexes SOD,GSH-Px,CAT,-SH and MDA in skeletal muscle was conducted using the reagent box.Groups b,c,B and D were trained by exhausting exercise and then were sacrificed immediately after being exhausted to prepare test samples and determine the above indexes.Results:After the exhausting exercise of swimming and treadmill,the anti-oxidant enzyme activity of skeletal muscle tissue was decreased greatly(p〈0.05) and the content of MDA produced by free radical metabolism was significantly increased(p〈0.05) in groups b and B.Compared with groups b and B,the anti-oxidant enzyme activity of skeletal muscle tissue of groups c and C was increased greatly(p〈0.05),the content of MDA produced by free radical metabolism was significantly decreased(p〈0.05),and the time of exercise to exhaustion was significantly prolonged.Conclusion:Exogenous gromwell extract supplement can improve the balance of free radical metabolism and prolong the exhaustive time of endurance-trained mice and rats.
