摘要 目的:micro RNA(mi RNA)是一类新的用于诊断疾病的生物标志物。目前关于吸烟与血浆mi RNA表达谱的研究相对较少。本研究通过血浆mi RNA表达谱芯片检测,筛选与吸烟相关的特异性mi RNA,为吸烟与健康研究提供一个新的方法。方法:应用人mi RNA表达谱芯片检测了28位青年吸烟者和12位非吸烟者血浆mi RNA表达谱,筛选了差异表达的mi RNA。应用生物信息学方法对差异表达的mi RNA进行了功能富集分析。结果:与非吸烟者比较,吸烟者血浆中35个mi RNA发生了差异表达。其中24个mi RNA表达上调,11个mi RNA表达下调。功能富集分析提示差异表达的mi RNA与免疫调节、细胞凋亡等生物学功能以及白血病等疾病密切相关。结论:吸烟可导致血浆mi RNA表达谱发生变化。差异表达的mi RNA可能成为吸烟与健康研究的新靶点。 Objective: Circulating micro RNA(mi RNA) is recognized as potential biomarkers of various diseases. This study aims to explore whether the level of circulating mi RNA in smoker vary with cigarette smoking by using plasma mi RNA expression profile to detect smoking-related specificity of mi RNA. Methods: In this study, the mi RNA expression profiles of 28 young smokers and 12 non-smokers were determined by Agilent human Micro RNA array. The enrichment function of differentially expressed mi RNAs induced by cigarette smoking was analyzed by bioinformatics method. Results: 35 mi RNAs were differentially expressed between smokers and non-smokers. 24 mi RNAs were up-regulated and 11 mi RNAs were down-regulated in the smokers. Bioinformatics analysis indicated that dysregulated mi RNAs was related to immune system and hormones regulation. Conclusion: Cigarette smoking changed plasma mi RNA expression profile in smokers. As a biomarker, mi RNA may help to understand the mechanisms by which disease responds to toxic substances included in tobacco smoking.
