雾化吸入布地奈德混悬液对哮喘患儿嗜碱性粒细胞活化状态影响研究 被引量:8

Effect of inhaled budesonide suspension on the activation of basophils in children with asthma

摘要 目的探究雾化吸入布地奈德混悬液对哮喘患儿外周血嗜碱性粒细胞活化状态的影响。方法收集2013年1月至2014年3月在上海交通大学医学院附属上海儿童医学中心就诊的哮喘患儿48例,作为哮喘组,以同期住院治疗的肺炎患儿53例为肺炎组,以儿保科门诊体检儿童49名为正常对照组。应用流式细胞术并结合粉尘螨刺激检测3组的外周血嗜碱粒细胞中CD63的表达,并进行比较;哮喘组雾化吸入布地奈德混悬液6个月后再次检测血嗜碱粒细胞中CD63的表达,对治疗前后进行比较。结果与肺炎组、正常对照组比较:粉尘螨刺激前,哮喘组嗜碱粒细胞CD63荧光强度显著增加(P<0.01);粉尘螨刺激后,哮喘组嗜碱粒细胞CD63荧光强度显著增加(P<0.01)。哮喘组在吸入布地奈德混悬液6个月后,粉尘螨刺激前,嗜碱粒细胞CD63荧光强度无变化(P>0.05);粉尘螨刺激后,嗜碱粒细胞CD63荧光强度下降(P<0.05)。结论嗜碱性粒细胞活化状态的检测可作为监测哮喘控制状态的参考指标,嗜碱粒细胞是吸入激素作用于儿童哮喘的重要靶标之一。 Objective To explore the effect of inhaled budesonide suspension on the activation of peripheral blood basophils in children with asthma. Methods Forty-eight children with asthma admitted between Januarry 2013 and March 2014 in Shanghai Children's Medical (;enter AMliated to Shanghai Jiao Tong University School of Medicine were adopted in asthma group. Fifty-three chihlren with pneumonia in the same period and 49 children performed physical examination in Department of Child Care were adopted in pneumunia group and normal enntrol group, respectively. Flow cytometry combined with dust utile stimulation was used In measure the expression uf CD63 in blood basophils of the three groups, and the resuhs were compared. The asthmalie ehihh'en were detected the expression of CD63 in blood basophil again after 6 monlhs of therapy with hndesonide suspension, artd the results were compared hefore and after treatment. Results Compared with the pneumonia group and normal control group, the hasophils fluorescence intensity of CD63 was increased significantly in asthma group before and after dust mite stimulation (P 〈 0.01 ). The asthmatic children receieved 6 months of trealment with inhaled budesonide suspension. The basophils flunreseenee intensity of CD63 did not ehange hefore dust mite stimulation (P〉 0.05) , but decreased significanlly after dust mile stimulation (P〈0.05). Conclusion The detection of basophil activation can be used to monitor the status ofasthma controrol The basophil is targets for inhaled corticosteroid asthma.
