摘要
目的探讨2型糖尿病(T2DM)对骨代谢的影响及其可能机制。方法收集C57BLKS/J-m/Lepr^(db)小鼠(A组,6只)和C57BL/6小鼠(B组,6只)的胫骨,采用Micro-CT和HE染色分析小鼠胫骨微结构,Western blot法检测骨形态发生蛋白2(BMP2)、BMP7、骨钙素(OCN)、Ⅰ型胶原(ColⅠ)和Runx2的蛋白表达。结果与B组相比,A组小鼠骨密度、骨小梁体积分数、骨小梁数及骨小梁厚度降低,骨小梁间隔增加,BMP2、OCN、BMP7、ColⅠ和Runx2蛋白表达均下调(P<0.01或P<0.05)。结论 T2DM可促使小鼠骨质疏松的发生,可能与其抑制骨形成作用有关。
Objective To investigate the effect and underlying mechanism of type 2diabetes mellitus(T2DM)on bone metabolism.Methods The tibiae of six C57BLKS/J-m/Leprdb mice(group A)and six C57BL/6 mice(group B)were collected.The microstructure of the tibia was analyzed by micro-CT and HE staining.The protein expressions of bone morphogenetic protein 2(BMP2),BMP7,osteocalcin(OCN),typeⅠ collagen(ColⅠ)and Runx2 were detected by Western blot.Results Compared with group B,the ratio of bone volumn to trabecular volumn,trabecular number,trabecular thickness and bone mineral density were decreased,trabecular separation was increased,and protein expressions of BMP2,OCN,BMP7,ColⅠ and Runx2 were down-regulated in group A(P〈0.01 or P〈0.05).Conclusion T2 DM can promote the development of osteoporosis,which may be related with the suppression of bone formation.
