月之汤饮对实验大鼠血液流变性及凝血功能的影响

Impact of the blood rheology and coagulation effect of yuezhitangyin decoction functional food on experimental rats

目的探讨月之汤饮对急性血瘀证模型大鼠血液流变性、正常大鼠血栓抑制率及凝血功能的影响。方法雌性成年大鼠,随机分为6组,即月之汤饮低、中、高剂量(2.5、5.0、10.0 g·kg-1)组、空白组、模型组、复方丹参片组(0.33 g·kg-1剂量灌服复方丹参片混悬液),各治疗组分别灌胃相应的药物,共7 d,皮下注射盐酸肾上腺素加冰水浴法复制大鼠血瘀证模型,观察月之汤饮对急性血瘀证模型大鼠血液流变性的影响。正常大鼠同法给药,共7 d,手术测定动静脉旁路血栓质量及抑制率影响,测定凝血时间。结果与模型组比较,月之汤饮5.0 g·kg-1剂量组的活化部分凝血酶原时间(APTT)较模型组显著延长(P<0.05),月之汤饮2.5、5.0、10 g·kg-1剂量组的凝血酶原时间(PT)较模型组显著延长(P<0.05),月之汤饮2.5 g·kg-1剂量组的血浆纤维蛋白原(FIB)含量较模型组显著增加(P<0.05);月之汤饮5.0、10.0 g·kg-1剂量组的血栓干、湿重量显著低于空白组(P<0.05或P<0.01)。月之汤饮2.5、5.0、10.0 g·kg-1剂量组均能显著延长大鼠凝血时间(P<0.01),凝血时间分别为25.20、21.11、20.10 s。结论月之汤饮能够抑制血栓形成,改善急性血瘀证模型大鼠血液流变性参数,具有活血化瘀的功效。

Objective To explore the thrombosis inhibition rate and bh）od cuagulation function of yuezhitangyin decoction on normal rats,and hemorheology on acute blood stasis model rats. Methods Aduh female rats were randomly divided into normal group,model group, positive eontrol group （ danshen tablets, dosing at 0.33g·kg^-1, respectively） and three yuezhitangyin deeoction groups （ dosing at 2.5,5,10.0g·kg^-1 ,respectively）. Each group was intragastrically administered corresponding drugs fi）r 7 d, and then subeutane- ously injected adrenaline and cool with ice water. Then the blood stasis and effects of yuezhitangyin decoction on the hemorheology in acute blood stasis model rats was determined. Each group was also intragastrieally administered corresponding drugs for 7 d to ohserve effects on thrombosis of normal rats by Yuezhitangyin decoction,and to determine its inhibition rate and the blood coagulation functiot）. Results Compared with the model group,APTT of Yuezhitangyin decoction （5.0 g·kg^-1） prolonged significmltly（ P 〈 0.05 ） , PT of Yuezhitangyin decoction （2.5,5.0,10.0 g·kg^-1 ） prolonged significantly （ P 〈 0.05 ） , FIB of Yuezhitangyin decoction （ 2.5 g·kg^-1 ） increaseded significantly（P 〈 0.05 ）. The wet and dry thrombus of all Yuezhitangyin decoction groups were runlet blank group weight （P 〈 0.05 or （P 〈 0.01 ）. The clotting time of tested drug groups exceeded blank group （ P 〈 0.01 ）, and the Y uezhitangyin decoction（2.5,5.0,10.0 g·kg^-1 ） were 25.20,21. 11 and 20. 10 s. Conclusions Yuezhitangyin deeoctiun has the effect of pronloting blood circulation and inhibiting blood stasis,can inbihit the thrombus forming and improve the blood rheology of acute blood stasis model rats.
