榄绿粗叶木茎甲醇提取物对CCl_4致小鼠肝损伤的保护作用 被引量:5

摘要 目的研究榄绿粗叶木茎甲醇提取物(简称MESLL)对CCl4引起肝损伤小鼠的保护作用。方法小鼠称重后随机分为6组。Ⅰ组为正常对照组,Ⅱ组为肝损伤模型组,均按20 m L·kg-1剂量每日一次性灌胃生理盐水;Ⅲ、Ⅳ和Ⅴ组分别按50、100、150 mg·kg-1剂量每日一次性灌胃MESLL溶液;Ⅵ组为按100 mg·kg-1剂量每日一次性灌胃水飞蓟素水溶液,连续7 d。末次给药2 h后,除正常对照组外,其他各组分别按10 m L·kg-1的剂量给小鼠一次性腹腔注射浓度为0.2%的CCl4溶液。禁食不禁水18h后,称重,摘眼球取血,测定血清AST和ALT活性。断颈处死小鼠,取其肝脏,称重。取肝脏右叶同一部位经10%甲醛固定,切片,HE染色,观察肝组织病理学变化。剩余肝脏组织匀浆,测定其T-AOC、SOD、MDA活性。结果各剂量的MESLL均能降低肝损伤小鼠血清ALT和AST水平,提高肝脏T-AOC、SOD活性和能降低MDA含量,减轻肝组织损伤程度。结论 MESLL对CCl4致小鼠急性肝损伤具有明显的保护作用。 Objective To evaluate the protective effect of methanol extract from the stem ofLasianthus lancilimbus (MESLL) on carbon tetrachloride (CCl4)-induced hepatic damage mice. Methods The mice were randomly divided into 6 groups: Group Ⅰ and Ⅱ were the normal control group and the model group, and they were both given 20 mL · kg - 1 normal saline. Group Ⅲ, Ⅳ and V were given MESLL at 50, 100 and 150 mg · kg- 1. Group Ⅵ were given 100mg· kg - 1 silymarin. All groups were given medicines or normal saline via intragastric administration once daily for 7 days. Two hours after the last intragastric administration, all the mice were given 0.2% CCl4 at 10 mL · kg - 1 except for the normal control group. After 18 h fasting, all the mice were weighed and blood samples were collected by eye bleeding method. The activities of ALT and AST of the blood samples were evaluated. Following the sacrifice of all the mice, the livers were dissected. The activities of SOD, MDA and T-AOC of the liver supernatant were tested. For histological analysis, the livers were fixed in 10% formaldehyde at room temperature. Then 5 μm thick serial sections were cut with microtome, and stained with haematoxylin-eosin. Results MESLL significantly reduced the level ofALT, AST and MDA, increased the level of T-AOC and SOD, and alleviated the liver injury in sinusoidal spaces in CCl4-induced hepatic mice. Conclusion MESLL can protect against hepatic injury in CCl4- induced mice.
