牡丹皮中丹皮酚的提取及包合工艺研究 被引量:8

摘要 目的优化牡丹皮中丹皮酚的提取及包合工艺。方法以丹皮酚提取率为指标,采用水蒸气蒸馏法,以温浸时间、加水量、蒸馏时间为考察因素,利用正交设计优化牡丹皮的提取工艺;采用饱和水溶液法,以丹皮酚包封率与包合物含药量为指标,以β-环糊精(β-CD)与丹皮酚摩尔比、搅拌时间、包合温度为考察因素,利用正交设计优化丹皮酚的β-CD包合工艺。结果牡丹皮中丹皮酚水蒸气蒸馏法最佳提取工艺是:加12倍量水,温浸2 h,蒸馏提取4 h,此提取工艺下丹皮酚的提取率为92.18%。丹皮酚β-CD最佳包合工艺是:β-CD与丹皮酚摩尔比为1.2:1,包合温度40℃,包合时间3 h,此包合工艺下丹皮酚包合率为72.38%,包合物含药量为7.94%。结论水蒸气蒸馏法是提取牡丹皮中丹皮酚较为适宜的方法,通过优化提取工艺可以获得理想的丹皮酚提取率。利用β-CD对丹皮酚进行包合,能有效提高其稳定性,通过优化包合工艺可以获得较高的丹皮酚包封率和包合物含药量。 Objective To optimize the extraction and inclusion of paeonol in Paeonia suffruticosa Andr.. Methods Orthogonal design was used to optimize the steam distillation extraction with paeonol yield as the index. The soaking time, water addition amount and distillation time were taken as study factors. With paeonol encapsulation efficiency and drug content as the indexes, the inclusion of paeonol with fl-cyclodextrin (β-CD) was studied by orthogonal design and the molar ratio offl-CD and paeonol, stirring time and temperature taken as study factors. Results The optimum steam distillation extraction of paeonol was 12 times of water, soaking for 2 h and distilled extraction for 4 h. The extraction rate of paeonol was 92.18%. The optimum inclusion for ofpaeonol with β-CD was the mole ratio of β-CD and paeonol at 1.2 : 1, mixing in 40℃ and inclusion for 3 h. The inclusion rate ofpaeonol was 72.38% and drug content was 7.94% under this inclusion process. Conclusion Steam distillation is a suitable extraction method for paeonol with good extraction rate. The stability of paeonol can be improved effectively by inclusion with β-CD as the inclusion material with good encapsulation efficiency and drug content.
