摘要
目的本研究通过RT-PCR和免疫组化方法检测间隙连接蛋白Cx26和Cx43在不明原因不孕组与对照组患者子宫内膜中的表达情况,比较Cx26和Cx43在子宫内膜组织中增殖期和分泌期的表达量以及是否Cx26和Cx43的表达不足与不明原因不孕症有关。方法采用免疫组化二步法和反转录PCR(RTPCR)技术,检测51例不明原因性不孕患者(不孕组)和45例继发性不孕患者(对照组)的增殖期及分泌期的Cx蛋白及其基因的表达情况。结果 (1)Cx26在不孕组与对照组的表达差异无统计学意义(P>0.05));(2)Cx43在对照组的表达高于不孕组,差异有统计学意义(P<0.05)。结论检测Cx43能够为不孕症的诊断提供依据,对其治疗提供有效的手段,而Cx26在此的意义不大。
Objective Investigated the variety of two connexins（Cx）expression in endometrium at proliferative phase and secretory phase.We also intended to find out the function of Cx in embryo implantation and development,as well the relationship between Cx and infertility.Methods（1）By using two-step immunohisto chemistry and RT-PCR,we detected Cx expressing in 51 cases of unexplained infertility（infertility group）and 45 cases of secondary infertility（control group）at proliferative phase and secretory phase.Results（1）The expression of Cx26 in the difference between the infertility group and the control had no significance（P〈0.05）;（2）The expression of Cx43 in the control group more than in the infertile group（P〈0.05）.Conclusion Study on the expression of Cx43 can be provided beneficial clues for infertility diagnosis and treatment,but Cx26 have no means in this side.
