六月雪水提取物体外清除羟自由基活性的实验研究 被引量:7

摘要 目的:探讨六月雪水提取物体外清除羟自由基活性,证明六月雪水提取物胃黏膜损伤的保护作用是否与清除自由基有关。方法:基于抗坏血酸还原Cu2+为Cu+,Cu+还原H2O2形成羟自由基·OH,·OH氧化邻菲罗啉,使其处于激发态,退激发时发光。抗氧化剂可以清除·OH,可以减少激发态邻菲罗啉的产生,使发光强度降低,运用此技术检测六月雪水提取物体外清除羟自由基的能力。结果:在六月雪水提取物的原浓度(3 000.000mg·m L-1)时,羟自由基被清除了98.978%,随着六月雪水提取物溶液的稀释,其清除羟自由基的能力逐渐降低,当稀释为原浓度的1/16 384浓度(0.183 mg·m L-1)时,六月雪水提取物溶液清除羟自由基的能力与空白组比较接近。结论:六月雪水提取物具有良好的体外清除羟自由基活性的能力,随着六月雪水提取物溶液的稀释,其清除羟自由基的能力逐渐降低。 Objective: To study the function of water extraction in Serissa serissoides in vitro to scavenge the activities of hydroxyl free radical,and too prove thatthe protective effect of Serissa serissoides water extract on gastric mucosa injury is related to scavenging free radicals. Methods: Cu2 +was reduced to Cu+by ascorbic acid,which was in the excited state,and the luminescence of the excitation state. Antioxidants which can scavenge ·OH,can decrease the excited state orth ophenanthroline and the emission intensity was employed to detect the scavenging ability of Serissa serissoides water extraction to hydroxyl radical in vitro. Results: With Serissa serissoides water extract of original concentration( 3 000. 00 mg·m L- 1),hydroxyl radical was cleared of 98. 978%. With the dilution of Serissa serissoides water extract solution,its ability to scavenge hydroxyl free radical decreased gradually. When the concentration of the dilution was the 1 /16 384 of the origional concentration( 0. 183 mg·m L- 1),the ability of Serissa serissoides water extract solution to scavenge hydroxyl free radical is close to that of the blank group. Conclusion: Serissa serissoides water extract has a good ability to savange the activity of hydroxyl free radical in vitro. As the dilution of the water extract solution in Serissa serissoides,its ability to scavange hydroxyl free radicals is gradually reduced.
