Apoptosis of Sinoatrial Node Cells in Spontaneously Hypertensive Rats

摘要 目的:观察自发性高血压大鼠窦房结(SAN)结细胞的凋亡,探讨厄贝沙坦对其影响。方法:将实验动物雄性大鼠(Wistar-Kyoto,WKY)和SHR随机分为:1WKY组(对照组)10只;2SHR组(模型组)10只;3厄贝沙坦治疗组(治疗组)10只(厄贝沙坦0.6 m L·100 g-1·d-1灌胃)。WKY组和SHR组同等剂量的生理盐水每天灌胃1次,持续喂养8周。SAN组织行TUNEL染色法,检测SAN细胞凋亡指数的变化。结果:WKY组SAN内可见散在的凋亡细胞,多分布于窦房结中央,在SHR组,可见SAN内凋亡细胞明显增加,呈不均一分布,凋亡细胞数量多,集中在窦房结中央;虽然治疗组凋亡细胞数量比SHR组减少,但仍比WKY组多见。结论:自发性高血压大鼠SAN 8周后出现结内细胞凋亡明显增加;自发性高血压大鼠行厄贝沙坦降低血压后可减少结内凋亡细胞发生。 Objective: To investigate cell apoptosis of Sinuatrial node and the intervention role of irbesartan in spontaneously hypertensive rats. Methods: WKY and spontaneously hypertensive rats were divided into three groups: 1 WKY group( control group),10 rats; 2 SHR group( the model group),10 rats; 3 irbesartan treated group( the treatment group),10 rats. The treated group were injected irbesartan 0. 6 m L ·100 g- 1·d- 1intragastric administration for 8weeks,while SHR group and WKY group were vehicle groups treated with the same dose saline. Paraffin sections of SAN were stained by TUNEL and detected the changes of apoptosis index. Results: Apoptosis cells were sporadic in the center of SAN in WKY group,whereas apoptosis cells in SAN in SHR group were more greatly than that in treated group,decreasing gradually from center to perimeter. The number of apoptosis cells in treated group were more than that in WKY group. Conclusions: Apoptosis cells in node are significantly increased after 8w in spontaneously hypertensive rats. Apoptosis cell of node were reduced after the spontaneously hypertensive rats were treated by irbesartan.
