静态张应力对人牙周膜成纤维细胞Wnt10b及DKK1蛋白表达的影响

摘要 目的研究不同静态张应力作用下人牙周膜成纤维细胞(h PDLFs)Wnt10b和DKK1蛋白的表达。方法采用酶解组织块法培养h PDLFs。用加载装置,对h PDLFs施加不同的静态张应力。按加力时间不同将细胞随机分为2 h、4 h和6 h组,每组再按弹力形变不同分为0%、8%、12%和16%形变组,0%为对照组。Western blot技术检测不同时间点和不同形变率h PDLFs中Wnt10b,DKK1蛋白的表达。各组分别进行组内比较。结果在2 h、4 h、6 h组加力与未加力的h PDLFs比较,Wnt10b蛋白表达显著减少,DKK1蛋白表达显著增多。各组内随着形变率的增大,Wnt10b蛋白表达逐渐减少,DKK1蛋白表达逐渐增多。结论静态张应力影响h PDLFs中Wnt10b及DKK1蛋白的表达。 Objective To investigate the effects of different static tensile stress on the expression of Wnt10 b and DKK1 in human periodontal ligament fibroblasts( h PDLFs). Methods Enzyme digestion method was used to culture h PDLFs. The cultured cells were randomly divided into three groups accroding to the duratioin of loading( 2 h,4 h,6 h). Cells in each group were divided into four subgroups with different deformation( 0%,8%,12% and 16%). 0%deformation served as control. The expressions of Wnt10 b and DKK1 were detected by Western blot. One-way analysis of variance was used to evaluate differences between different deformations at each time point. Results In h PDLFs under static tensile stress,the expression of Wnt10 b was reduced significantly compared with the control group,while DKK1 expressed more heavily. At each time point,the expression of Wnt10 b was reduced with the increasing tension stress,while DKK1 was increased. Conclusion Wnt10 b and DKK1 may participate in the metabolism of h PDLFs. Static tensile stress inhibits the canonical Wnt / β-catenin signaling in h PDLFs.
