摘要
为了探讨SIRT1基因在猪卵巢颗粒细胞生殖激素分泌中的作用,采用白藜芦醇和尼克酰胺处理猪卵巢颗粒细胞,检测猪卵巢颗粒细胞中SIRT1基因和生殖激素受体表达水平,分析SIRT1基因与生殖激素受体表达量的相关性。结果表明,白藜芦醇和尼克酰胺可剂量依赖性地调节猪卵巢颗粒细胞中SIRT1基因表达,SIRT1基因表达量的升高可引起ER2、FSHR、LHR表达量上调,其相关系数分别为0.820 6、0.519 7和0.140 7。说明,SIRT1基因参与猪卵巢颗粒细胞生殖激素受体表达调控,可影响猪卵巢颗粒细胞生殖激素分泌。
To explore the role of SIRT1 gene in secretion of reproductive hormones in porcine ovarian granulosa cells,quantitative real-time PCR was employed to detect the m RNA expressions of SIRT1 gene and reproductive hormone receptors genes in granulosa cells treated with resveratrol and nicotinamide. The correlation between expression levels of SIRT1 and reproductive hormone receptors genes was also analysised by SPSS software. Results showed that expression of SIRT1 gene was regulated by resveratrol and nicotinamide in granulosa cells in a dose-dependent manner. With the upregulation of SIRT1 m RNA expression,m RNA expressions of ER2,FSHR,LHR were increased. Correlations between SIRT1 and ER2expressions,between SIRT1 and FSHR expressions and between SIRT1 and LHR expressions were positively linear,with the correlation coefficients of 0.820 6,0.519 7 and 0.140 7,respectively. These results suggest that SIRT1 may take part in the regulation of reproductive hormone receptors expressions,thereby affecting the secretion of reproductive hormones in porcine ovarian granulosa cells.
