Smad3基因调控食管鳞癌发生及EGCG干预研究 被引量:2

摘要 目的探讨Smad3基因与食管鳞癌发生及发展关系,表没食子儿茶素没食子酸酯(epigallocatechin-3-gallate,EGCG)抑制食管癌细胞Ec9706增殖作用及调控Smad3基因表达水平。方法 RT-PCR方法检测44例食管鳞癌及正常食管组织中Smad3基因表达水平,流式细胞术方法检测38例原发性食管鳞癌和20例正常食管组织的细胞周期,不同浓度EGCG(0、100、200、300 mg/L)作用Ec9706细胞24、48 h后流式细胞术检测细胞增殖及Smad3蛋白表达水平。结果食管鳞癌组织中Smad3 mRNA表达水平显著低于正常食管组织中的表达水平,差异有统计学意义(P<0.05)。食管鳞癌组织增殖指数显著高于正常食管组织(P<0.05)。不同浓度EGCG作用Ec9706细胞24 h后,细胞增殖指数显著降低(P<0.05),而Smad3蛋白表达水平显著增高(P<0.05)。结论 Smad3在食管鳞癌中的异常表达参与了食管鳞癌发生及发展,EGCG具有抑制食管鳞癌生长作用与调控Smad3蛋白表达有关。
