摘要
以赤霞珠葡萄(Vitis vinifera L.)为材料,以Actin基因为内参,采用半定量逆转录聚合酶链式反应(RT-PCR)法检测葡萄果实发育过程中白藜芦醇合酶(STS)基因表达量的变化,以期建立适于检测该基因表达的RT-PCR试验体系。结果表明,在退火温度58℃,扩增循环31次的时候,STS基因能够进行较好的扩增。在赤霞珠葡萄浆果发育的过程中,花后20、50 d STS基因表达量较少,在花后80 d表达量增到最大,花后110 d又降低。
With cabernet sauvignon(Vitis vinifera L.) grapes as test materials and Actin gene as an internal reference, the expression of resveratrol synthase(STS) gene in berries on grape fruit development process were detectined to establish an appropriate system of RT-PCR by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) method. The results showed that STS gene had a better amplification when the annealing temperature was equal to 58 ℃ and the number of amplification cycles was equal to 31 times. In the process of development of cabernet sauvignon berries, STS gene had a less expression on 20 days and 50 days after flowers, the maximum expression were found on 80 days after flowers, and then decreased on 110 days after flowers.
