摘要 目的:探讨注意缺陷多动障碍大鼠模型的建造技术。方法:将20只4周龄SD大鼠随机分成模型组和对照组,模型组自由饮用0.2 mg/m L醋酸铅溶液。28 d后运用开场实验、Morris水迷宫、Lat迷宫观察两组大鼠行为学表现;采用ICP-MS检测大鼠血铅浓度、酶联免疫吸附测定法法(ELISA)测定大鼠血清中多巴胺和去甲肾上腺素含量。结果:模型组在开场实验中大鼠穿越格子数、直立次数增多(P<0.05),Lat迷宫中直立次数及穿越角落的时间比例增加(P<0.05),Morris水迷宫目标象限停留时间缩短(P<0.05);模型组大鼠血铅浓度高于对照组(P<0.05),血清多巴胺和去甲肾上腺素含量低于对照组,差异有统计学意义(P<0.05)。结论:采用自由饮用0.2 mg/m L醋酸铅溶液法成功建造了ADHD大鼠模型,具有ADHD基本临床特征,可做为ADHD的动物模型进行实验研究。 Objective: To explore the method of constructing rat models of attention deficit hyperactivity disorder( ADHD). Methods: 20 SD rats were randomly divided into the model group and the control group with the model group drinking 0. 2 mg / m L solution of lead acetate. 28 days later,the rats' behavioral manifestations were observed by using the open field,the Morris water maze and the Lat maze; the rats' lead concentration in the blood was determined by using ICP- MS; the contents of dopamine and norepinephrine in the rats' serum were determined by using enzyme linked immune sorbent assay( ELISA). Results: The occasions of the rats' in the model group going through the lattice standing up were more and more frequent in the open field( P〈0. 05); the occasions of the rats' standing upright were more frequent and the time for the rats to go across the corner was lengthened in the Lat maze( P〈0. 05); the target quadrant time in the Morris water maze was shortened( P〈0. 05); the lead concentration in blood of the model group was significantly higher than that of the control group( P〈0. 05); the contents of dopamine and norepinephrine in the serum of the model group were lower than those of the control group,the difference being significant( P〈0. 05). Conclusion:Rat models of ADHD can be constructed successful by freely drinking 0. 2 mg / m L solution of lead acetate,which have the basic clinical characteristics of ADHD and can be studied as animal models of ADHD.
