摘要
核苷类似物是一类重要的抗肿瘤药物,对其进行结构改造修饰,提高药物的生物利用度,减少不良反应,已成为核苷类抗肿瘤新药的研究热点。本文通过对药物的化学结构进行分类,分别介绍了碱基修饰、糖基修饰、碱基和糖基同时修饰以及前药修饰的几类核苷类药物及其药理作用机理,阐述了近年来具有抗肿瘤活性的核苷类药物的最新研究进展,为核苷类抗肿瘤药物的研究与开发提供参考。
Nucleoside analogue is an important type of antitumor agents. Structure modification of existing drugs can not only improve the drug bioavailability but also reduce adverse reactions,and it has become a research hotspot of nucleoside anti-tumor agents. In this article,the nucleoside agents were classified according to chemical structure. Several types of drugs,including modification in bases,carbohydrates,both the base and carbohydrates and prodrug modification were reviewed respectively.
