脉复生通过核因子-κB通路对脂多糖诱导血管内皮细胞损伤的保护作用 被引量:5

Protective Effect of Maifusheng on Vascular Endothelial Cells from Injury Induced by Lipopolysaccharide Through Nuclear Factor-kappa B Pathway

摘要 【目的】观察脉复生(主要由牛大力、熟地黄、白花蛇舌草等组成)对脂多糖(LPS)诱导的人脐静脉内皮细胞(ECV304)内核因子-κB(NF-κB)信号通路蛋白表达的影响。【方法】体外培养ECV304细胞,实验分为正常对照组,LPS诱导组(终浓度为1μg/m L),LPS诱导+脉复生高、中、低剂量组(终浓度分别为10、1、0.1μg/m L),采用Western-blot法检测各组细胞中NF-κB p65、肿瘤坏死因子-α(TNF-α)、一氧化氮合酶(i NOS)、环加氧酶-2(COX-2)、血管细胞黏附因子-1(ICAM-1)及细胞间黏附分子-1(VCAM-1)的蛋白表达水平。【结果】与正常对照组比较,LPS诱导组细胞上清液中NF-κB p65、TNF-α、i NOS、COX-2、ICAM-1、VCAM-1蛋白表达均显著增加(P<0.01);脉复生各剂量组能不同程度地减少LPS诱导细胞中NF-κB p65、TNF-α、i NOS、COX-2、ICAM-1、VCAM-1蛋白表达水平,其中中、高剂量组作用显著(P<0.05或P<0.01)。【结论】脉复生可通过抑制LPS诱导的ECV304细胞中NF-κB、TNF-α、i NOS、COX-2、ICAM-1、VCAM-1的蛋白表达,调节NF-κB信号通路,从而发挥保护血管内皮细胞的作用。 Objective To observe the influence of Maifusheng on the expression of signaling pathway protein nuclear factor kappa B(NF- κB) in human umbilical vein endothelial cells(ECV304) induced by lipopolysaccharide(LPS). Methods In-vitro cultured ECV304 cells were divided into normal control group,LPS induction group(at final concentration of 1 μg/m L), and high-, middle- and low-dose Maifusheng groups(LPS 1 μg/m L plus Maifusheng at final concentration of 10,1 and 0.1 μg/m L,respectively). The expression levels of NF-κB p65,tumor necrosis factor-alpha(TNF-α),nitric oxide synthase(i NOS),cyclooxygenase-2(COX-2),intercellular adhesion molecule-1(ICAM-1) and vascular cell adhesion molecule-1(VCAM-1) in various groups were determined by Western blotting method. Results Compared with the normal control group,NF-κB P65,TNF-α,i NOS,COX-2,ICAM-1 and VCAM-1 protein expression levels were increased in the supernatant of LPS induction group(P〈0.01). The protein expression levels of NF-κB p65, TNF-α, i NOS,COX-2, ICAM-1 and VCAM-1 in Maifusheng groups were decreased to different degrees, the differences being significant in high- and middle-dose groups(P〈0.05 or P〈0.01 compared with LPS induction group).Conclusion Maifusheng plays a role in protecting vascular endothelial cells from injury through inhibiting the protein expression of NF-κB p65,TNF-α,i NOS,COX-2,ICAM-1 and VCAM-1,and regulating the NF-κB signaling pathway.
