摘要
目的探讨转化生长因子(TGF)-β1-869T/C,TGF-β_1-509C/T基因单核苷酸多态性(SNPs)与鼻咽癌易感性与复发的关系。方法采用病例对照研究,采集来自辽宁地区不同医院的鼻咽癌患者193例,同期就诊的非肿瘤患者231例静脉血,酚氯仿法提取基因,Taqman real-time聚合酶链反应(PCR)方法及SDS软件对TGF-β_1基因型进行分型。SPSS 13.0软件包进行数据分析,以χ~2检验比较SNPs基因型在病例与对照之间分布的差异,以单因素和多因素Logistic回归计算比值比(OR)及95%可信区间(CI)。结果单因素分析显示,TGF-β_1-869位点的各基因型与鼻咽癌易感性不相关,同样TGF-β_1-509位点的各基因型与鼻咽癌易感性也不相关,经校正年龄性别吸烟状态后显示仍与鼻咽癌易感性仍不相关。经单因素及多因素进一步分析后显示:TGF-β_1-869、TGF-β_1-509位点各基因型与鼻咽癌复发也不相关,差异无统计学意义。结论 TGF-β_1-869和TGF-β_1-509位点SNPs与鼻咽癌易感性及复发均不相关。
Objective To investigate the association of TGF-β_1 gene single-nucleotide polymorphisms with susceptibility and recurrence of nasopharyngeal carcinoma.Methods One hundred and ninty-three historically diagnosed and followed nasopharyngeal carcinoma patients and 231 healthy people were included.After collecting the peripheral blood samples for all,single nucleotide polymorphisms were analyzed by Taqman real-time PCR.Genotypes of all samples were analyzed with SDS program.All statistical analyses were performed with statistical product and service solutions v13.0.ORvalue and 95% confidence interval(CI)were calculated.Results Single factor analysis showed that TGF-β_1-869 genotype has no relationship with the susceptibility of nasopharyngeal carcinoma;the same as the result of TGF-β_1-509 genotype,there was no relationship with susceptibility of nasopharyngeal carcinoma either.Multiple factors analysis considering age,gender and smoking also showed that both TGF-β_1-869 and TGF-β_1-509 genotype didn′t increase the risk of nasopharyngeal carcinoma significantly.After further univariate and multivariate analysis,we didn′t find any association between every genotype of TGF-β_1-869 and TGF-β_1-509 and recurrence of nasopharyngeal carcinoma,Pvalue has no significant difference.ConclusionTGF-β_1 genes polymorphism have no relation with the susceptibility and recurrence of nasopharyngeal carcinoma.
