矢车菊色素对高龄糖尿病大鼠B细胞淋巴瘤基因2及Bcl-2相关X蛋白基因表达的影响

摘要 目的了解矢车菊色素对高龄2型糖尿病大鼠(Goto-Kakizaki rats,GK大鼠)肾损害时B细胞淋巴瘤伯血病基因-2(B cell lymphoma/lewkmia-2,Bcl-2)、Bcl-2相关X蛋白(Bcl-2 assaciated X protem,Bax)基因表达的影响。方法 25周龄雄性GK大鼠24只随机分为矢车菊组和对照组,每组12只。矢车菊组饲喂含10%葡萄糖的矢车菊色素-3-葡萄糖苷溶液600 mg/L8周,对照组饲喂10%葡萄糖溶液8周。每周测量体重及空腹血糖,8周后处死。RT-qPCR检测大鼠左肾Bcl-2、Bax mRNA表达,同时检测左肾脏器指数、血清糖化血红蛋白(glycated hemoglobin A1c,GHbA1c)并观察右肾病理损害。结果矢车菊组肾Bcl-2 mRNA表达高于对照组,差异有统计学意义(P<0.05)。矢车菊组Bax基因与对照组比,mRNA的表达差异无统计学意义(P>0.05)。矢车菊组GK大鼠的体重、空腹血糖、血清GHbA1c和左肾脏器指数较对照组均降低(P<0.05)。与对照组比较,矢车菊组肾脏病理损害明显减轻。结论矢车菊色素可降低GK大鼠的体重、空腹血糖和血清GHbA1c水平,减轻糖尿病的肾损伤,可能与促进Bcl-2基因表达有关。 Objective To investigate the influence of cyanidin-3-glucoside on gene expression of Bcl-2 and Bax in kid- ney lesion of old GK rats. Methods 24 healthy male GK rats were divided randomly into two groups: b]ank control group (n = 12) and cyanidin group (n = 12). cyanidin group was fed in 10% glucose with cyanidin-3-glucoside (600 mg/L). The blank control group was fed in 10% glucose only. The weight, fasting plasma glucose (FPG) were detected each week. 8 weeks later, all GK rats were killed. The mRNA expression of Bcl-2 and Bax was detected by RT-qPCR. The left kidney weight index, serum GHbAlc and histopathologicalchange of right kidney were examined too. Results The mRNA ex- pression of Bcl-2 was higher than the blank control group (P〈0.05). No statistical significance of the Bax mRNA expression were detected between the cyanidin group and blank control (P〉 0.05). The weight, FPG, serum GHbAlc and left kidney weight index in cyanidin group were lower than the blank control group ( P〈 0.05 ). The renal? pathologic lesion in cyanidin group was lighter than the blank control group. Conclusion The weight, FPG, GHbAlc and kidney diabetes lesion of GK rats can be decreased by cyanidin-3-glucoside perhaps through the increasing of Bcl-2 mRNA expression.
