防己黄芪汤对阿霉素肾病大鼠肾组织乙酰肝素酶表达的影响 被引量:11

摘要 目的观察防己黄芪汤对阿霉素肾病大鼠肾小球滤过功能的作用,探讨该方减少蛋白尿,治疗肾小球疾病的可能机制。方法尾静脉注射阿霉素法建立阿霉素肾病大鼠模型,成模后,根据24 h尿蛋白定量随机分为模型组、防己黄芪汤高、低剂量治疗组,连续给药5周,分别测定各组大鼠24 h尿蛋白定量及肾组织乙酰肝素酶(HPA)表达。结果模型组大鼠24 h尿蛋白定量和肾组织HPA表达升高明显高于正常组(P<0.05);与模型组比较,防己黄芪汤高、低剂量治疗组24 h尿蛋白定量和肾组织HPA表达均明显改善(P<0.05)。结论防己黄芪汤减少模型大鼠尿蛋白的作用可能与调节肾组织HPA表达,保护足细胞受损有关。 Objective: To study the protection of Fangji Huangqi Decoction on the Adriamycin Induced Nephropathy(AIN) rats and explore its mechanism. Methods: Wistar rats were given intravenous injections of ADR via tail vein,establishing adriamycin-induced nephropathy rat model. The model rats were randomly divided into the model group and the treatment groups of high-dosage and low-dosage medicine,and were then administered intragastrically by medicine in corresponding dosage. In next 5 weeks,rats in each group were detected in24 hours urinary protein. Renal tissues routine was fixed,and heparanases expression was detected with reverse transcription PCR. Result: Those indices in the modle group were higher obviously than those in the routine group including the 24 hours urinary protein and the renal tissue HPA. However,these indicators of the high-dosage and low-dosage medicine group were improved signifcantly than the modle group(P〈 0.05). Conclusion:Fangji Huangqi Decoction can reduce the quantity of 24 hours urine protein of Adriamycin Induced Nephropathy rats,and protect the glomerular filtration barrier by moderating the expression of heparanases.
