摘要
目的观察益气活血方的急性毒性试验以及对大鼠慢性脊髓压迫后受损脊髓组织内超氧化物歧化酶(SOD)和丙二醛(MDA)含量的影响。方法 1)20只ICR小鼠随机选取5只为假手术组,余15只为实验组,采用最大给药量的方法观察益气活血方的急性毒性反应。2)60只SD大鼠,随机选取10只作为假手术组,仅暴露椎板,未造成脊髓压迫,以生理盐水进行灌胃;其余的SD大鼠采用自体第7颈椎棘突椎间移植法制造大鼠脊髓压迫模型。术后1 d根据脊髓功能的评分将其分为益气活血方组:造模后,以不同浓度的益气活血方进行灌胃。颈复康组:造模后,以颈复康进行灌胃。模型对照组:造模后,以生理盐水进行灌胃。连续灌胃4周后,采用ELISA方法测定大鼠受损节段中SOD和MDA的含量。结果小鼠的最大给药量相当于生药的892.5 g/(kg·d),为临床成人日用量的315倍,在此剂量下未见明显毒性反应,7 d内无1例动物死亡;不同浓度的益气活血方及颈复康颗粒均能显著增加大鼠受损脊髓组织内SOD的含量(P<0.05);中药高、中浓度以及颈复康颗粒均能显著降低MDA的含量(P<0.05),中药低浓度也降低MDA的含量,但是与模型对照组相比较差异无统计学意义(P>0.05)。结论益气活血方是安全的且能有效提高受损脊髓组织细胞中氧自由基的清除率,促进脊髓功能的恢复。
Objective: To study the acute toxicity test and the potential effects exerted on the contents of SOD and MDA in damaged rats′ spinal cord tissue of the Yiqi Huoxue Decoction. Methods: 1)20 ICR mice were randomly selected,5 for the control group,the rest for the experimental group,and then the method of maximum dosage was adopted to observe the acute toxicity response of PNQAB. 2)10 SD-Rats were chosen as the shamoperated group randomly from the sixty subjects,fed with the normal saline within explosion of the vertebral lamina and freed from oppression of the spinal cord;the remaining were applied to establish the rat′s spinal oppression model by grafting the self 7th cervical vertebrae spinous process at the part of C4-5 or C5-6 intervertebral space,then divided into three groups based on the assessment results of the spinal cord function at the 1st day postoperatively,fed with the Yiqi Huoxue Decoction under different concentrations(Group A),Jinfukang(Group B) and Normal saline(Group C),respectively. After treating 4 weeks,all the rats were narcotized to get the specific spinal cord for the determinations of SOD and MDA. Results: The daily maximum dosage equal to crude drugs to mice was 892.5 g/kg,315 times of daily clinical dosage of adult. Within the dose,obvious toxic effects was not seen,ang no animals died within 7 days. The results showed that the Yiqi Huoxue Decoction regardless of the concentration difference and Jingfukang Particles could both increase the content of SOD in rats′ damaged spinal cord tissues significantly(P〈 0.05);while the Yiqi Huoxue Decoction within high/medium concentrations and Jingfukang particles could also notably decrease MDA content,but not for the Yiqi Huoxue Decoction in low concentration. Conclusions: The Yiqi Huoxue Decoction is safe and can effectively enhance the scavenging rate of the reactive oxygen radicals in tissues of the damaged spinal cord,and promotes the recovery of the spinal cord function.
