锡类散结肠溶胶囊对DSS诱导小鼠结肠炎的干预作用研究

The interference of Xilei San Jiechangrong capsule on DSS-induced ulcerative colitis in mice

目的:探讨锡类散结肠溶胶囊对葡聚糖硫酸钠(DSS)诱导小鼠溃疡性结肠炎的疗效及可能作用机制。方法:BALB/c雄性小鼠40只,随机分为正常组、模型组、对照组、治疗组,10只/组。除正常组饮用普通的饮用水外,其余各组小鼠饮予含5%DSS的饮用水7d,复制溃疡性结肠炎模型。对照组给予柳氮磺吡啶0.4g·kg-1·d-1、治疗组给予锡类散结肠溶胶囊,正常组、模型组给予同体积生理盐水,连续10d。观察小鼠结肠重量/单位长度比值变化,疾病活动度指数(DAI)及病理组织学损伤情况,采用免疫组化技术检测结肠黏膜肿瘤坏死因子(TNF-α)、白细胞介素(IL-10)含量。结果:治疗组与对照组小鼠腹泻、便血情况明显好转,DAI评分均低于模型组;模型组小鼠结肠重量/单位长度比值与正常组、对照组、治疗组比较有显著性差异;经病理学观察,正常组小鼠结肠黏膜皱襞纹理清晰,无炎细胞浸润,模型组小鼠结肠可见充血水肿,伴有局部糜烂、溃疡,有较多炎细胞浸润,而治疗组与对照组小鼠组织学损伤均有所缓解,结肠充血水肿及炎细胞浸润现象减轻,溃疡面减少;与模型组比较,对照组、治疗组TNF-α水平明显降低,而IL-l0水平明显上调。结论:具有结肠定位释放效应的锡类散结肠溶胶囊对DSS诱导的小鼠溃疡性结肠炎呈现较好的治疗效应,能明显缓解模型小鼠炎性症状。

Objective： To investigate efficacy of Xilei San Jiechangrong capsule on dextran sulphate sodium induced ulcerative colitis in mice. Methods： Forty male BALB/c mice were randomly divided into four groups： the normal group, the DSS model group, the Salazosulfapyridine control group, the Xilei San Jiechangrong capsule treatment group. Except for mice in the normal group, all mice were administered with 5% DSS solution, for 7 days to induce colitis. From 8th to 17 th day, the normal group and the DSS model group were given normal saline by gavage daily; the SASP control group and the Xilei San Jiechangrong capsule treatment group were administered with SASP 0.4g·kg-1·d-1 and the Xilei San Jiechangrong capsule by gavage daily respectively. The body weight and stool consistency and occult or gross blood were recorded to calculate the disease activity index. The ratio of colon weight and length were evaluated. The macroscopic and histopathological changes of the mice colon were observed. The level of TNF-α and IL-l0 were assessed by immunohistochemical method. Results： Compared with the DSS model group, the SASP control group and the Xilei San Jiechangrong capsule group significantly ameliorated the symptom of UC and histopathological, reduced colonic DAI, decreased the levels of colonic TNF-α and increased the levels of colonic IL-10. Conclusion： The Xilei San Jiechangrong capsule demonstrated good effects on ulcerative colitis induced DSS. It provided an experimental basis to the research and development of Xilei San Jiechangrong capsule.
