参附注射液对血管性痴呆模型大鼠学习记忆能力及神经细胞凋亡的影响 被引量:7

摘要 目的:探讨参附注射液对血管性痴呆(Va D)模型大鼠学习记忆能力及神经细胞凋亡的影响。方法:采用Pulsinelli四血管阻断法(4-VO)制作Va D大鼠模型,将36只大鼠随机分为假手术组、模型组和参附注射液组。采用Morris水迷宫检测学习记忆能力,采用免疫组化染色检测海马区Bax、Bcl-2、Caspase-3阳性表达。结果:参附注射液组大鼠逃避潜伏期缩短,跨越平台次数增加,与模型组比较差异均具有统计学意义(P<0.01)。参附注射液组大鼠海马区Bcl-2阳性细胞表达显著增多,而Bax和Caspase-3阳性细胞表达明显减少,与模型组比较差异均具有统计学意义(P<0.01)。结论:参附注射液能够通过下调凋亡相关基因Bax和Caspase-3的表达,上调抗凋亡基因Bcl-2水平,发挥抗海马神经元凋亡作用,改善Va D大鼠学习记忆能力。 Objective To explore efficacy of the Shenfu injection on learning and memory capacity and neuronal apoptosis in vasculardementia rat models. Methods: Vascular dementia rat models were made by 4-VO. 36 rats were randomly divided into the sham group, modelgroup and Shenfu injection group. Learning and memory capacity was tested by Mossis water maze. Bax, Bcl-2, Caspase-3 in hippocampuswere tested by immunohistochemistry. Results: Rats in the Shenfu injection group showed shorter escape latency and gained more activity, withstatistically significant difference when compared with the model group (P〈0.01). Bcl-2 positive cells in hippocampus of rats was significantlyincreased, and Bax, Caspase-3 positive cells and significantly reduced, with statistically significant difference compared with the model group(P〈0.01). Conclusion: By reducing the expression of Bax and Caspase-3, and improving the level of anti-apoptotic Bcl-2 genes, the Shenfuinjection played anti-hippocampal neuronal apoptosis and improved learning and memory capacity roles in rats VaD.
