兔VX2门静脉癌栓模型构建 被引量:4

The establishment of VX2 portal vein tumor thrombus model in experimental rabbits

摘要 目的 经肠系膜上静脉插管注入VX2肝癌瘤粒或瘤条,构建不同类型兔门静脉癌栓(PVTT)模型。方法 剖腹后直视下穿刺肠系膜上静脉,在兔门静脉主干近端置入经皮肝穿刺胆道造影(PTC)套管。20只健康大白兔随机分成两组,A组(n=10)注入0.1-0.2 cm瘤粒,B组(n=10)注入0.1 cm×1.5-2.0 cm瘤条。术后1周,DSA造影观察癌栓生长情况,处死后作病理学观察。结果 两组实验兔均成功完成门静脉插管,瘤株接种成功率均为100%。术后1周造影显示,A组8只兔见门静脉一级分支癌栓,2只兔见门静脉主干癌栓,10只兔均见肝内弥漫散在的多发转移灶;B组10只兔均见门静脉主干癌栓,肝实质内未见弥漫性转移灶。结论 采用介入技术将不同大小的VX2肿瘤瘤粒或瘤条种植于兔门静脉内,能形成伴有或不伴有肝内弥漫性转移灶的门静脉主干或一级以上分支癌栓模型。 Objective To establish different types of portal vein tumor thrombus (PVTr) models in rabbits by injecting VX2 tumor particles or strips through superior mesenteric vein catheterization. Methods After exploratory laparotomy, superior mesenteric vein was punctured under direct vision, then, a percutaneous transhepatic cholangiography (PTC) cannula was placed in the proximal side of main portal vein. Twenty healthy rabbits were randomly divided into group A (n=10) and group B (n=10). Particles of 0.1-0.2 cm sized VX2 tumor tissue were injected into portal vein in the rabbits of group A, while 0.1× (1.5-2.0) cm sized tumor tissue strips were injected into portal vein in the rabbits of group B. One week after the implantation, digital subtraction angiography (DSA) was performed to observe the growth of PVTr. Results Technical success of portal vein catheterization was achieved in all rabbits of both groups, the success rate of tumor strain vaccination was 100%. DSA performed one week after the implantation showed that in group A PVTT was formed in the primary branch of portal vein (n=8) or in the main portal vein (n=2), and multiple disseminated hepatic metastases were demonstrated in all 10 rabbits; while in group B the formation of PVTT in the main portal vein was observed in all the 10 rabbits, and no disseminated liver metastases were seen. Conclusion By using interventional technology, the implantation of different sized VX2 tumor particles or strips into the portal vein can establish rabbit models of PVTT, locating in the main portal vein or in the primary branch of portal vein accompanying with or without multiple disseminated hepatic metastases.
