摘要
目的:保障提取总RNA质量的前提下探索一种保存样品的适宜方法。方法:Trizol法(试剂盒)提取缺血复灌后肾组织总RNA。缺血45分钟后再灌注的大鼠肾脏,按照不同保存方法将样品分成四组,第一组液氮处理后立即提取总RNA;第二组-20℃冻存1天后提取总RNA;第三组-20℃冻存1周后提取总RNA;第四组液氮处理后1周后提取总RNA,然后检测各组RNA含量。结果:第一组RNA含量最多,第三组提取总RNA的含量最少。结论:提取总RNA的肾组织样品最佳保存方法是获得组织后立即提取总RNA,或者经液氮处理后保存在-20℃,一周后仍可获得较多含量的总RNA。
Purpose: looking for a good method to store the sample which can get the high quality total RNA. Methods: total RNA extraction( Trizol kit) from rat ischemic kidney. The kidney cut from the rat after ischemia / reperfusion 45 min,samples store for four groups: 1. the RNA extracts by total RNA extraction kit immediately after the sample treated by liquid nitrogen; 2. the RNA extracts by total RNA extraction kit one day later and the sample store at-20℃; 3. the RNA extracts by total RNA extraction kit one week later and the sample store at-20℃; 4. the RNA extracts by total RNA extraction kit one week later and the sample store at liquid nitrogen. Result: Group one is the most and the group three is the least. Conclusion: extract the total RNA immediately is the best method,or week later after the sample store at-20℃ with liquid nitrogen treating,will still obtain more total RNA.
