CnIH侧脑室注射后大鼠下丘脑组织中神经肽Y mRNA及蛋白表达变化 被引量:2

摘要 目的观察促性腺激素抑制激素(Gn IH)侧脑室注射后大鼠下丘脑组织中神经肽Y(NPY)mRNA及蛋白表达变化。方法雄性SD大鼠60只,随机分为实验组和对照组各30只,分别给予侧脑室注射Gn IH(1μg/μL,5μL)和生理盐水(5μL),分别于注射1、2、4 h后各处死10只,取下丘脑组织。采用real-time PCR法检测NPY mRNA,免疫印迹法检测NPY蛋白。结果实验组注射后1、2、4 h下丘脑组织中NPY mRNA相对表达量分别为0.759±0.025、1.755±0.219、2.671±0.696,NPY蛋白相对表达量分别为0.225±0.015、0.244±0.015、0.264±0.009;对照组分别为0.633±0.021、0.568±0.006、0.631±0.015和0.113±0.004、0.125±0.007、0.128±0.011。两组注射后2、4 h NPY mRNA表达量相比,P均<0.05;两组注射后各时点NPY蛋白表达量比较,P均<0.05。结论Gn IH侧脑室注射后,大鼠下丘脑组织中NPY mRNA及蛋白表达上调。
