三邻甲苯磷酸酯对Neuro-2a细胞生长分化的影响

摘要 目的研究三邻甲苯磷酸酯(Tri-ortho-cresyl phosphate,TOCP)对Neuro-2a(N2a)细胞生长分化的影响,探索TOCP诱导迟发性神经病(OPIDN)发生的机制。方法培养N2a细胞,利用CCK8试剂盒检测TOCP抑制细胞增殖的浓度范围,然后分别以0、2.5、5、10和20μmol/L浓度的TOCP染毒N2a细胞,观察TOCP对视黄酸(Retinoic acid,RA)诱导N2a细胞分化的影响。在此基础上,利用TOCP染毒分化的N2a细胞,观察TOCP对N2a细胞轴突形态的影响。结果TOCP染毒引起未分化的N2a细胞分化能力显著减弱,轴突长度明显缩短;而分化的N2a细胞暴露于TOCP后,则出现明显的轴突损伤,且随着TOCP染毒剂量的增加,轴突损伤逐渐加重。结论 TOCP影响了N2a细胞轴突的生长分化,可能与其诱导OPIDN发病机制有关。
