摘要
目的研究血管生成素-2(angiopoietin-2,Ang-2)在高氧诱导的小鼠视网膜新生血管动物模型中的表达,探讨Ang-2在视网膜新生血管形成中的作用。方法高浓度氧诱导C57BL/6J小鼠建立视网膜缺血性病变动物模型。将20只新生小鼠随机分为高氧诱导组和正常对照组。将高氧诱导组小鼠置于氧气浓度为(75±2)%的密闭容器中饲养,5 d后将幼鼠取出置于正常环境中饲养,正常对照组幼鼠置于正常环境中饲养。两组幼鼠均于生后17天处死,用免疫组化和Real-time PCR的方法检测幼鼠视网膜Ang-2蛋白及mRNA表达情况。结果高氧诱导组幼鼠突破内界膜的新生血管内皮细胞数为25.68±2.61,明显高于正常对照组1.45±1.01,两组比较差异有统计学意义(P<0.05)。高氧诱导组幼鼠Ang-2 mRNA的相对表达量是3.01±1.11,明显高于正常对照组1.47±0.53,两组比较差异有统计学意义(P<0.05)。两组的Ang-2平均光密度值分别是121.25±8.61和75.45±7.87,两组比较差异有统计学意义(P<0.05)。结论高氧诱导组幼鼠眼球中Ang-2表达明显增加,视网膜新生血管的形成显著增多,提示Ang-2在视网膜新生血管形成过程中可能发挥着重要作用。
Objective To investigate the expression of angiopoietin-2(Ang-2)in the retina of hyperoxia-induced model of retinopathy and to evaluate the effect of Ang-2 in retinal neovascularization. Methods C57BL/6J mice were put into the environment of high oxygen to establish models of ischemic retinopathy. Twenty mice were randomly divided into hyperoxic and normoxic control groups. The hyperoxic group mice were put into closed container where oxygen concentration was 75±2% for 5 days,then moved into normal air. The normoxic group mice were raised in normal air. Mice were sacrificed in postnatal day 17,the expression of Ang-2 was examined in the mice retina using immunohistochemistry and reverse transcription-polymerase chain reaction.Results The neovascular endotheliocyte was higher in hyperoxic group(25.68±2.61)than which in normoxic group(1.45±1.01)(P〈0.05). The m RNA expression of Ang-2 was higher in hyperoxic group(3.01±1.11)than which in normoxic group(1.47±0.53)(P〈0.05). The optical density value of Ang-2 was higher in hyperoxic group(121.25±8.61)than which in normoxic group(75.45±7.87)(P〈0.05). Conclusion The retinal neovascularization and the Ang-2 expression were increased in hyperoxic group,suggesting that Ang-2 may related to retinal neovascularization.
