摘要 目的:研究阿片生长因子Met-ENK(methionine enkephalin,Met-ENK)对多种肿瘤细胞的抑制作用,分析其对不同肿瘤细胞系的抑制作用差异性及特点。方法:采用CCK-8法考察不同浓度Met-ENK对SH-SY5Y细胞的增殖抑制活性;采用细胞计数法计数相同浓度Met-ENK作用下,SHSY5Y细胞随时间的生长曲线;采用CCK-8法检测不同浓度的Met-ENK对8种不同的肿瘤细胞系细胞的抑制作用。结果:不同浓度的Met-ENK对SH-SY5Y细胞具有不同的抑制作用,IC50为5.1×10^(-5)mol/L;Met-ENK作用后SH-SY5Y在48 h生长抑制明显(P<0.01);不同浓度的MetENK对8种不同的肿瘤细胞系细胞均有抑制作用,其中对SMCC-7721抑制作用最为明显。结论:Met-ENK具有广谱抗肿瘤作用,但抑制强度不同,具有时间依赖性,其对8种不同的肿瘤细胞系细胞均有不同强度的抑制作用。 AIM: To study the inhibition effect of Met-ENK on a wide variety of tumor cells,analyzing its differences of inhibition effect on different tumor cell lines. METHODS: CCK 8 was used to investigate the different concentrations of Met ENK to inhibit the growth of SH SY5Y. Count method was used to draw the curve that the SH-SY5Y growth inhibition as changes over time under the same concentration. Different concentrations of Met-ENK were adopted on eight kinds of tumor cells by CCK 8 method. RESULTS: The inhibition effects of different concentrations of Met-ENK on SH-SY5Y cell line were different. At different time points especially in 48 hours Met-ENK obviously inhibited the growth of SH-SY5Y( P 〈0. 01). Different concentrations of Met-ENK had inhibitory effects on eight tumor cells especially SMCC-7721. CONCLUSION: Met-ENK has broad-spectrum inhibition effect on tumor cells,the inhibitory effect is timedependent. Met-ENK has inhibition effects on eight different tumor cell lines with various degrees.
