氧甾酮133和重组人骨形态发生蛋白-2对兔后外侧腰椎模型中腰椎融合率的比较 被引量:3

Effect of oxysterol 133 vs. recombinant human bone morphogenetic protein -2 on fusion rate in a rabbit posterolateral lumbar spine model

摘要 目的探讨氧甾酮133(Oxy133)和重组人骨形态发生蛋白-2(rhBMP-2)对兔后外侧腰椎模型融合率的影响。方法对24个月龄雄性成年新西兰白兔(3.0~3.5kg)的L3-L4脊椎实行双侧后外侧脊椎融合术。实验兔分成4组:对照组(A组)、30txgrhBMP-2(B组)、20mgOxy133(C组)、60mgOxy133(D组)。4周后,荧光显影检查其融合,8周后处死兔,通过放射显影、临床触诊和计算机断层扫描技术评估其融合效率。结果8周时放射显影技术进行融合率分析各组分别为:A组41.7%;B组91.7%;C组91.7%;D组100.O%。处死兔后剥离其脊柱通过临床触诊分析其融合率结果:A组0.0%;B组83.3%;C组83.3%;D组66.7%。计算机断层扫描结果进一步确定以上结果。结论20mg和60mgOxy133都能促进骨融合,融合效率基本与30斗grhBMP-2接近,均显著高于对照组,表明Oxy133有希望替代rhBMP-2成为促进骨融合的治疗剂。 Objective To compare the effects of recombinant human bone morphogenetic protein- 2 (rhBMP- 2) vs. oxysterol (Oxy)133 on fusion rate in a randomized controlled trial using a posterolateral lumbar rabbit spinal fusion model. Methods Twenty - four male adult white New Zealand rabbits (3.0 - 3.5 kg) underwent bilateral posterolateral lumbar spinal fusion at L4 - LS. Rabbits were di- vided into four groups: control (A), 30 -mg rhBMP -2 (B), 20 - mg Oxy 133 (C), and 60 - mg Oxy 133 (D). At 4st week, fusion was evaluated by fluoroscopy, and at 8th week, the rabbits were sacri- riced and fusion was evaluated radiographically, by manual palpation, and with microcomputed tomo- graphy. Results Radiographic analysis showed the rusion rate at 8th week in groups A, B, C and D was 41.7%, 91.7% , 91.7%, and 100.0% respectively. Evaluation of fusion masses by manual palpation of excised spines after sacrifice showed the fusion rate in group A, B, C and D was 0. 0%, 83.3% , 83.3% and 66.7% respectively. Microcomputed tomography scanning confirmed these findings. Conclusion These findings in a rabbit model demonstrate that both 20 - and 60 - mg Oxy 133 doses promote fusion that is equivalent to fusion induced by 30 - mg rhBMP - 2 and significantly greater than the control group. The present findings confirm that Oxy 133 is a promising candidate for therapeutic development as an alternative to rhBMP- 2 to promote spinal fusion.
