摘要
目的观察人胎盘源间充质干细胞(PMSC)联合外周血造血干细胞(PBSC)共移植入严重联合免疫缺陷(SCID)小鼠体内后对其早期造血的影响。方法将人胎盘组织经胶原酶消化、贴壁和传代培养获得人PMSC,运用流式细胞仪(FCM)检测其表面标志;以人PMSC和PBSC为供体,通过尾静脉注射的方法,将两种细胞单独或联合移植入经亚致死量照射预处理的SCID小鼠体内,18只受鼠随机分为3组,每组6只:A组为生理盐水对照组;B组为PBSC移植组;C组为PMSC与PBSC联合移植组。移植后的28d取各组存活SCID小鼠股骨中的骨髓细胞,并用FCM检测分析人CD34^+、CD45^+细胞的植入水平。结果从人胎盘组织中成功分离和培养PMSC,FCM检测其表型正常;SCID小鼠移植后28d,对照组小鼠存活1只,造血干细胞组和联合移植组小鼠6只全部存活,C组股骨骨髓腔内的人CD34^+、CD45^+细胞的百分比[(39.06±2.23)%、(36.54±1.63)%]均明显高于B组小鼠。结论人PMSC可以促进PBSC的植入,缩短造血干细胞移植后造血恢复时间。
Objective To investigate the effect of transplantation of human placenta mesenchymal stem cells (PMSCs) combined with peripheral blood stem cells (PBSCs) on early hematopoietic stage in severe combined immune deficiency (SCID) mice. Methods We obtained human PMSCs from human placental tissue by collagenase, digestion, adherent culture and subculture and detected surface markers by flow cytometry. PBSCs from donated human peripheral blood were transplanted alone or combined with PMSCs into sublethally irradiated SCID mice via tail -vein injection. Eighteen recipient SCID mice were randomly divided into there groups: group A (the saline control group), group B (PBSCs transplantation group), and group C (the human PMSCs combined with PBSCs transplantation group). At day 28 after transplantation, we achieved femur bone - marrow cells from survival SCID mice in each group and detected the levels of human CD34^+ and CD45^ + by flow cytometry (FCM) analysis. Results At day 28 after trans- plantation, the human PMSCs from placental tissue were normal by FCM analysis. Only one mouse survived in control group A, and all mice survived in group B and group C. The proportion of CD34^+ and CD45^+ cells with transplantation of both cell types in the femur medullary cavity [ (39.06 ± 2. 23 )% and (36. 54 ± 1.63) % ] was higher than in PBSCs alone. Conclusion Human PMSCs can promote the im- plantation of PBSCs, and shorten the hematopoietic recovery time of hematopoietic stem cell transplanta- tion.
