剑麻皂素对2型糖尿病(T2DM)大鼠的治疗作用及其机制探讨

摘要 目的:探讨剑麻皂素对T2DM大鼠的治疗作用及其作用机制。方法:高糖高脂饮食加小剂量链脲佐菌素(STZ)联合诱导制备T2DM大鼠模型。研究剑麻皂素对T2DM大鼠血脂、血糖、胰岛素、氧化应激反应、血管内皮功能、主动脉功能的影响。结果用药治疗12周后,剑麻皂素能明显降低糖尿病大鼠血清血糖(GLU)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、游离脂肪酸(FFA)、丙二醛(MDA)、血管性血友病因子(v WF)、糖基化终末产物(AGEs)含量;提高高密度脂蛋白(HDL-C)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSHPx)、一氧化氮(NO)、胰岛素(INS)含量;同时能改善糖尿病大鼠离体主动脉的收缩和舒张反应。结论:剑麻皂素具有调节T2DM大鼠血糖、胰岛素、血脂、主动脉功能等作用,其作用机制可能与改善机体的氧化应激和血管内皮功能有关。 Objective: To investigate the effects and mechanism of Tigogenine on streptozotocin-induced diabetic rats. Methods: High glucose and high fat diet, combined with small dose streptozotocin was used to establish type 2 diabetes rat mode. The changes of blood lipids, blood glucose, insulin release, oxidative stress,vascular endothelium and thoracic aorta in rats were observed. Results: After Tigogenine treatment for 12 weeks,the contents of blood GLU, TC, TG, LDL-C, FFA, MDA, v WF, AGEs in rats obviously decreased, and the levels of HDL-C, SOD, GSH-Px, NO, INS significantly increased. Tigogenine also improved the vasodilation and vasoconstriction function. Conclusion: Tigogenine has positive effects on the blood glucose, insulin, blood lipids and aortic function in T2 DM rats, which may due to its anti-oxidative and endothelial protective effects.
