摘要
【目的】探讨碱性成纤维细胞生长因子(bFGF)、血管内皮细胞生长因子(VEGF)浓度梯度对于猪体外血管内皮细胞、成纤维细胞增殖移行的影响。【方法】以不同浓度VEGF、bFGF刺激猪血管内皮细胞、成纤维细胞,筛选出在各时间点细胞增殖及移行活力均有显著差异的三个浓度构成浓度梯度。以胶原膜片加栽VEGF、bFGF,以MTT法和计算细胞迁移距离法比较不同时间点浓度梯度组与单一浓度组细胞增殖移行活力的差异。【结果]VEGF浓度梯度组在24h、48h、72h时间点,VEGF浓度梯度组OD值均显著小于对照组25ng/mL和100ng/mL(P〈0.05)IVEGF浓度梯度组6h、12h、24h迁移距离明显大于对照组5ng/mL组、25ng/mL(P〈0.05),但6h、12h、24h与对照组100ng/mL比较无显著性差异(P〉0.05),在12h小于对照组100ng/mL(P〈0.05)。在不同时间点bFGF浓度梯度组0D值与对照组10ng/mLOD值比较无显著性差异(P〉0.05);但均小于对照组50ng/mL和100ng/mL(P〈0.05);bFGF浓度梯度组在不同时间点的迁移距离均显著大于对照组10ng/mL、50ng/mL、100ng/mL,其差异均有统计学意义(P〈0.05)。【结论]VECF、bFGF浓度梯度对于血管内皮细胞、成纤维细胞的迁移活力促进作用高于单一浓度,但因受生长因子浓度影响,而对于血管内皮细胞、成纤维细胞的增殖活力较单一低浓度无明显促进作用。
[Objective] To study the impact of bFGF and VEGF concentration gradient on porcine vascular endothelial cells and proliferation of fibroblast migration.[Methods]Porcine vascular endothelial cells and fibroblasts were stimulated with different concentrations of VEGF and hFGF, and three concentrations at each time point were selected when cell proliferation and migration activity were significantly different so as to form a concentration gradient. VEGF and bFGF were loaded with collagen membrane. The difference among different time points of concentration gradient group and single concentration group of proliferation activity were compared by the MTT method and the calculation of cell migration distance method.[Resuits] After 24 h, 48 h and 72 h, the proliferative activity expressed by OD value in VEGF Concentration gradient group was significantly less than that Of the control group 25 ng/mL and 100 ng/mL ( P %0.05) . However, at 6 h,12 h and 24h , there was no significant difference compared with the control group of 100 ng/mL ( P 〉0.05), which was less than that of the control group 100 ng/ mL at 12 h ( P 〈0.05) . At different time points, OD value of bFGF concentration gradient group showed no significant difference with that of the control group 10 ng/mL ( P 〉0.05), but less than the control group of 50 ng/ mL and 100 ng/mL ( P 〈0.05) ; BFGF concentration gradient group migration distance in different time points were significantly higher than those of the control group 10 ng/mL, 50 ng/mL, 100 ng/mL. The differences were statistically significant ( P 〈0.05).[Conclusion] The promoting effect of migration activity of VEGF and bFGF concentration gradient for vascular endothelial cells and fibroblasts is higher than that of the single concentration, but due to the influence of concentration of the growth factor, it has no obvious role in promoting proliferation activity for vascular endothelial cells and fibroblast comparing with a single low concentration.
