ADAM10、E-cad及flt-1在重度子痫前期胎盘中的表达及意义 被引量:6

The expression and the roles of a disintegrin,metalloproteinase 10 and its'zymolyte in severe preeclampsia placenta

摘要 目的通过对孕妇胎盘组织中解聚素金属蛋白酶10(a disintegrin and metalloproteinase 10,ADAM10)及其底物E-钙黏蛋白(epithelial cadherin,E-cad)、底物受体血管内皮生长因子受体-1(like tyrosine kinase-1,Flt-1)的检测,探讨三种因子与子痫前期发病的关系。方法采用RT-PCR和Western Blot方法分别检测39例重度子痫前期组和39例正常妊娠组胎盘组织中三种因子mRNA和蛋白的表达。结果重度子痫前期组ADAM10和Flt-1mRNA表达(1.7347±0.1705和1.1325±0.0823)均高于正常妊娠组(1.0512±0.0932和0.8254±0.0752),其蛋白质水平也均高于正常妊娠组(P<0.001)。而两组E-cad mRNA(0.3277±0.0251,0.5996±0.0450)和蛋白表达(0.4789±0.1148和0.7601±0.1260)低于正常妊娠组(P<0.001)。结论重度子痫前期胎盘组织中ADAM10、Flt-1过度表达及E-cad低表达可能与子痫前期的发生和发展相关。 Objective To detect the expression of a disintegrin and metalloproteinase 10(ADAM10)and its' zymolyteepithelial cadherin(E-cad)and the receptor of zymolyte like tyrosine kinase-1(Flt-1)and in the placenta and study its relationship with the pathology of preeclampsia.Methods 39 cases of severe preeclampsia and 39 cases of nomalpregnanceas control were enrolled in the study.The placental tissues were obtained after delivery.The expression of three factor mRNA in the placental tissues was detected by RT-PCR method and the protein was detected by Western blot method.Results The expression of ADAM10 and Flt-1in mRNA and proteinlevel were significantly higher in severe preeclampsia group than that in control group,while E-cad was lower in severe preeclampsia group(P〈0.001).Conclusion The over-expression of ADAM10 and Flt-1and the lower-expression of E-cad in the placenta of sever preeclampsia patients may be associated with the occurrence and development of preeclampsia.
