摘要
目的探讨低频电刺激对人宫颈癌SiHa细胞的影响。方法对处于生长对数期宫颈癌SiHa细胞,根据低频电刺激强度分为20mA、40mA和60mA及对照组,刺激时间30mins,采用CCK-8增殖试验检测细胞的增殖能力。采用Transwell小室进行人工重组基底膜侵袭和转移试验,检测细胞侵袭和转移能力。结果不同低频电刺激后,四组4、24、48、72h细胞增殖力比较,差异无统计学意义(P>0.05)。Transwell小室试验显示,不同低频电刺激后,四组细胞侵袭转移力比较,差异无统计学意义(P>0.05)。结论低频电刺激对人宫颈癌SiHa细胞的增殖和侵袭转移能力无明显影响,但尚需进一步研究证实。
Objective The aims of this study is to investigate the effects of electrical stimulation on human cervical cancer line-SiHa cells.Methods The experimental parameter was designed according to the low frequency electrical stimulation parameter for the treatment of radical hysterectomy patients with urinary retention.According to the different current intensity,the electrical stimulation group was divided into group 20 mA,group 40 mA and group 60 mA.Group no stimulation is the control.The electric stimulation was 30 mins for the group 20 mA,group 40 mA and group 60 mA respectively.The proliferation ability was measured with CCK-8proliferation assay.The invasion and immigration capacity was evaluated with transwell cave method.Results After different current intensity stimulation,There was no significant difference in cell proliferation among the four groups(P〈0.05).there was no significant difference in cell invasion and metastasis(P〈0.05)after different stimulation among the three groups and the control.Conclusion This study indicated that low frequency electrical stimulation has no significant effect on the proliferation and invasion of SiHa cell line.But it needs further study afterward.
