摘要
目的:探讨癃畅颗粒治疗良性前列腺增生症的作用机制。方法:120只Wistar大鼠随机原则分为6组,即癃畅颗粒低剂量组、中剂量组、高剂量组、癃闭舒组、模型对照组、正常组,除正常组外,其余组大鼠采用去势后注射丙酸睾酮的方法造模。造模的同时,治疗组每天灌胃给药1次,模型组灌以等量生理盐水,连续30天,采用TUNEL法检测前列腺组织细胞凋亡状况,IHC法检测前列腺组织细胞Survivin的表达状况。结果:与模型组比较,癃闭舒组和癃畅颗粒各剂量组大鼠前列腺细胞凋亡指数均有不同程度升高,Survivin表达率不同程度降低(P<0.01);癃畅颗粒高剂量组与癃闭舒组比较差异无统计学意义(P>0.05);癃闭舒组与癃畅颗粒中剂量组、低剂量组比较差异有统计学意义(P<0.01);癃畅颗粒不同剂量组间比较差异有统计学意义(P<0.01或P<0.05)。结论:癃畅颗粒可以下调大鼠前列腺增生组织中Survivin的表达,促进细胞凋亡,这可能是其治疗前列腺增生症的作用机制。
Objective: To investigate the mechanism of Longchang granules in the treatment of benign prostatic hyperplasia. Methods: Divide 120 Wistar rats into 6 groups: normal group,model group,Longbishu group,low dose Longchang granule group,medium dose Longchang granule group and high dose Longchang granule group. Except normal group,all of the rest rats were made into models of prostate hyperplasia by testosterone propionate injection after they were first castrated. All the model rats were killed after treated respectively with Longchang granules,Longbishu and normal saline by gastrogavage once a day for 30 days. The apoptosis of cells was identified by TUNEL assay. IHC method was applied to detect the expression of Survivin gene in prostate tissue. Results: Compared with model group,the prostate apoptosis indexes of all treatment groups were all increased to different levels,while the expression of Survivin protein reduced to deferent levels( P 0. 01). No statistical significance could be found between high dose Longchang granule goup and Longbishu group( P 0. 05). While a statistical significance could be found between medium,low dose Longchang granule groups and Longbishu group( P 0. 01). There was a statistical significance among all groups of Longchang granules( P 0. 01). Conclusion: Longchang granules can downregulate the expression of Survivin gene in prostate tissue of benign prostate hyperplasia rats and accelerate cell poptosis. This is the possible mechanism of Longchang granules in the treatment of benign prostatic hyperplasia.
