摘要
目的:建立测定山药丸中士的宁和马钱子碱含量的方法。方法:采用HPLC法,色谱柱为Luna C18(4.6 mm×250 mm,5μm);流动相为乙腈-0.01 mol·L^(-1)庚烷磺酸钠与0.02 mol·L^(-1)磷酸二氢钾等量混合溶液(用10%磷酸调节p H值至2.8)(21∶79),流量1.0 m L·min^(-1),柱温35℃,检测波长为260 nm。结果:士的宁和马钱子碱的线性范围分别为0.03~0.95μg(r=1.000 0)、0.03~1.17μg(r=0.999 5),平均回收率(n=6)分别为98.5%、99.0%,其RSD分别为1.4%、0.8%。结论:所建方法可靠、准确,可用于该制剂的质量控制。
Objective:To establish a method for determining Strychnine and Brucine in Shanyao Pills. Methods: HPLC method was adopted ona Luna C18 column(4.6 mm × 250 mm,5μm) with acetonitrile- 0.01 mol· L^-1 sodium heptanesulfonate and 0.02 mol · L^-1 potassium dihydrogen hosphate mixed solution (adjusted pH to 2. 8 with 10% phosphoric acid) (21: 79) as mobile phase. The flow rate was 1.0 mL · min^-1, column temperature was 35 ℃ and detection wavelength was 260 nm. Results:The linear range of Strychnine and Brucine was 0. 03 -0.95 μg(r = 1. 000 0) and 0.03 - 1.17 μg(r =0. 999 5), respectively. The average recovery rate of Strychnine and Brucine was 98.5 % and 99.0% ( n = 6) with RSD of 1.4% and 0. 8%, respectively. Conclusion: The method is reliable and accurate, which can be used for the quality control of the preparation.
