铁皮石斛碱溶性和水溶性多糖的比较研究 被引量:6

摘要 目的建立鲜铁皮石斛的碱溶性和水溶性多糖柱前衍生HPLC特征图谱,并初步比较其单糖组成及活性的差异。方法采用高效液相色谱法测定,比较铁皮石斛2种多糖的1-苯基-3-甲基-5-吡唑啉酮(PMP)-HPLC特征图谱、单糖的组成、相似度。应用WST-1细胞增殖及细胞毒性检测试剂盒(WST-1法)测定2种多糖的抗氧化活性,根据其清除羟自由基(·OH)、过氧化氢(H2O2)的能力,评价其抗氧化活性。结果铁皮石斛水溶性多糖的单糖组成以D-甘露糖和D-葡萄糖为主;碱溶性多糖的单糖组成以D-甘露糖、D-半乳糖醛酸、D-葡萄糖、D-半乳糖和D-木糖为主。生物活性初步研究表明铁皮石斛碱溶性多糖和水溶性多糖均具有一定的抑制Hela细胞增殖的作用。铁皮石斛碱溶性多糖清除羟自由基(·OH)、过氧化氢(H_2O_2)的能力均强于水溶性多糖。结论铁皮石斛碱溶性和水溶性多糖的特征图谱、生物活性均有差异,而引起两者活性差异的原因可能跟单糖组成有关。 Objective To establish the HPLC characteristic spectra of water-soluble and alkali-soluble polysaccharides from D. officinale and analyze the difference of the monosaccharide composition and bioactivity. Methods The PMP-HPLC fingerprints of both polysaccharides were established,then the difference of their spectra,monosaccharide composition and similarity were analyzed. The WST-1 method was used to evaluate the anti-tumor activity of polysaccharides. The antioxidant activity of polysaccharides were evaluated according to the ability of clearing · OH and H2O2. Results The water-soluble polysaccharides were mainly composed of D-mannose and D-glucose,while alkali-soluble polysaccharides contained D-mannose,galacturonic acid,D-glucose,D-galactose and D-xylose. The bioactivity studies demonstrated that both alkali-soluble and water-soluble polysaccharides could inhibit the proliferation of Hela cells. However,the antioxidant activities of alkali-soluble polysaccharides were more significant than the water-soluble polysaccharides. Conclusion There are differences between alkali-soluble and watersoluble polysaccharides on the characteristic spectra and activities,which may be caused by the different monosaccharides compositions.
