摘要
以四种不同品系大花君子兰的幼子房为外植体进行了离体组织培养研究.结果表明:大花君子兰幼子房愈伤诱导和分化成苗的能力与品系、外植体时期、光照强度、低温处理时间、植物生长调节剂种类和质量浓度及酸水解酪素质量浓度等有关.愈伤诱导的最佳培养基为MS+蔗糖35g/L+酸水解酪素0.4g/L+BA 1.0mg/L+NAA 1.0mg/L+2,4-D 2.0mg/L;分化成苗的最佳培养基为MS+蔗糖35g/L+NAA 0.5mg/L+KT 3.0mg/L;生根的最佳培养基为1/2 MS培养基.
A tissue culture system responsible for four strains of Clivia miniata Regel was developed using young ovaries as explants.The results showed that callus induction and differentiation rate was influenced by cultivar background,status of explant,light intensity,temperature treatment,components and concentration of plant growth regulators as well as concentration of casein hydrolysate.The best callus induction medium was MS+sucrose 35g/L+hydrolysis casein 0.4g/L+BA 1.0 mg/L+NAA 1.0 mg/L+2,4-D 2.0 mg/L,the optimum regeneration medium was MS+sucrose 35g/L+NAA 0.5 mg/L+KT 3.0 mg/L,and 1/2 MS was showed to be the best rooting medium.
