摘要
[目的]探讨甘草酸二铵对大鼠体内卡马西平(CBZ)及其代谢产物10,11-环氧卡马西平(ECBZ)药物代谢动力学的影响.[方法]将SD雄性大鼠随机分为生理盐水(NS)对照组、甘草酸二铵中、高剂量组,各组分别腹腔注射给予NS、甘草酸二铵50,100mg/kg,每日1次,连续7d,于第8d腹腔注射给予NS、甘草酸二铵1h后,灌胃给予CBZ 50mg/kg.采用HPLC分析方法测定大鼠血浆中CBZ及ECBZ的含量,并利用WinNolin 5.2药物代谢动力学软件计算药物代谢动力学参数.[结果]与NS对照组比较,甘草酸二铵中、高剂量组CBZ与ECBZ的t1/2,CL/F,AUC0~12和Cmax等药物代谢动力学参数差异均无统计学意义(P〉0.05).[结论]在大鼠体内甘草酸二铵不影响CBZ的药物代谢动力学过程.
OBJECTIVETo study the effect of diammonium glycyrrhizinate(GLN)on the pharmacokinetics of carbamazepine(CBZ)and its metabolites 10,11-epoxy-carbamazepine(ECBZ)in rats.METHODS SD male rats were randomly divided into groups of CBZ + normal saline control(NS),CBZ+middles dose GLN and CBZ+high dose GLN.The NS and 50,100mg/kg GLN in each group were administrated by intraperitoneal injecton once a day for 1week,and then 50mg/kg CBZ were given by gavage after 1hof NS or GLN administration at the 8th day.The plasma concentrations of CBZ and ECBZ in rats were determined by HPLC,and the pharmacokinetic parameters were calculated by using WinNolin 5.2pharmacokinetic software.RESULTS As compared with CBZ+NS,CBZ+GLN group have no significant changes in pharmacokinetic parameters of CBZ and ECBZ(P0.05).CONCLUSION GLN has no effect on CBZ pharmacokinetics in rats.
