摘要
目的:探讨白细胞介素4(interleukin-4,IL-4)基因多态性与脑梗死间的相关性。方法:选取189例急性脑梗死患者(脑梗死组)和92例非急性脑梗死者(对照组),运用聚合酶链反应-限制性片段长度多态性(polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)技术,结合血液生化和血压等临床资料,对IL-4基因-590C/T和33C/T 2个位点的基因单核苷酸多态性与脑梗死间的关系进行研究。结果:IL-4基因-590C/T位点上,脑梗死组与对照组间的基因型、等位基因频率分布有显著差异(P<0.01);脑梗死组TT型患者的血糖、收缩压、舒张压均明显高于对照组(P均<0.05)。IL-4基因33C/T位点上,脑梗死组与对照组间的基因型、等位基因频率分布差异均无统计学意义(P>0.05);脑梗死组TT型患者的收缩压、舒张压参数均明显高于对照组(P<0.05),而其高密度脂蛋白胆固醇水平则明显低于对照组。结论:IL-4基因-590C/T多态性的TT型可能与我国上海地区人群的脑梗死发病相关。
Objective To explore the correlation of interleukin-4(IL-4) gene single nucleotide polymorphism(SNP)with the occurrence of cerebral infarction(CI). Methods: Blood samples were collected from 189 patients with acute CI and 92 controls. Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) assay was used to analyze the specific SNP loci of IL-4 gene(-590C/T, 33C/T). Combined with blood pressure and blood biochemistry, correla-tions of SNP loci of IL-4 gene(-590C/T, 33C/T) with CI were analyzed statistically. Results: The frequencies of genotype TT and allele(T) of IL-4-590C/T polymorphism in CI group were statistically different from those in control group( P〈0.01).Patients with TT genotype in CI group had higher glucose levels and blood pressure(P〈0.05). There were no statistically significant differences in distribution of genotypes and allele frequencies of IL-4 33C/T gene polymorphisms(P〉0.05) between CI group and the control group. However, CI patients with TT genotype had higher blood pressure and lower HDL-C level(P〈0.05) when compared with the control group. Conclusions: T allele of-590C/T polymorphism in IL-4 gene might be associated with an increased risk of CI in Shanghai population.
