辽砧2号组培苗瓶内生根技术研究

Study on Rooting Technology in Bottle of Apple Stock in Vitro of Liaozhen NO.2

以苹果矮化砧木辽砧2号组培苗为试材,研究不同培养条件对组培苗瓶内生根的影响。结果表明,辽砧2组培苗在以1/2MS为基本培养基、附加IBA 0.5 mg/L与NAA 0.3 mg/L、培养温度(25±1)℃、光照时间16 h/d、光照强度为2500 lx的条件下,培养40 d的生根率可达73.33%;生根培养基中加入一定量的活性炭,极大提高组培苗的生根率,可达86.67%。不但解决了生根难或生根慢问题,而且有利于壮苗,便于移栽。

The apple dwarf rootstock Liaozhen No.2 seedlings as experimental material,effect of different culture conditions on rooting in the bottle. The results showed that the 16h/d was the basic medium,and the addition of NAA 0.3 mg/L and 1/2MS,the culture temperature was（25 ± 1）℃,the light intensity was 2 500 Lx,the rooting rate of the 40 days was 73.33%. rooting of activated carbon by adding a certain amount of medium greatly increased the rooting rate of apple dwarf rootstock,the rooting rate was 86.67%.Not only solve the problem of slow or difficult rooting and rooting,but also conducive to the healthy,easy to transplant.
