不同灸法对实验性RA家兔关节滑膜组织HSP70表达的影响

Effect of Different Types of Moxibustion Treatment on Expression Levels of HSP70 in RA Rabbits

目的:观察3种灸法(温和灸、直接灸和隔姜灸)对实验性RA模型的抗炎效应,比较研究不同灸法对实验性RA家兔关节滑膜组织HSP70表达的影响。方法:40只家兔随机分为对照组、RA模型组、温和灸组、直接灸组、隔姜灸组,8只动物/组。注射FCA造模后第7 d开始分别用温和灸、直接灸、隔姜灸3种不同方法施灸家兔"足三里"、"肾俞"2穴,两侧交替进行,治疗1次/d,6 d为1疗程,共计3个疗程。于造模前、造模后7 d及治疗后7、14、21 d测量各组家兔双后肢膝关节周长,采用S-P免疫组化法检测滑膜组织HSP70的含量。结果:温和灸、直接灸和隔姜灸均能明显降低家兔左右膝关节周长(P<0.01),隔姜灸作用优于温和灸和直接灸(P<0.05);与对照组比较,RA模型组关节滑膜组织HSP70积分光密度值增高(P<0.05);与RA模型组比较,各艾灸组滑膜组织HSP70积分光密度值显著增加(P<0.01),隔姜灸组较温和灸组、直接灸组增加更为明显(P<0.05)。结论:不同灸法对实验性RA均有良好的抗炎效应,以隔姜灸作用更为突出;不同灸法均可诱导实验性RA家兔关节滑膜组织HSP70大量表达,以隔姜灸更为明显。

Objective： To observe the effect of different moxibustion intervention on the inflammatory reactions and the expression of HSP70 in the synovium of hind-knee joints in rheumatoid arthritis（ RA） rabbits. Methods： 40 Japanese white rabbits were randomized into 5 groups（ n = 8） ： normal control,RA model,mild-warm moxibusiton,direct-moxibustion,and ginger-partitioned moxibustion groups. RA model was injected by Freud＇s Complete Adjuvant（ 0. 5 m L / kg） into the articular cavities of rabbits＇ bilateral hind-limbs. Moxibustion treatment was given to unilateral ＂Zusanli＂（ ST 36） and ＂Shenshu＂（ BL 23） points alternatively from the 7thday on after modeling,once daily for 3 weeks except Sundays. The circumference of the hind-knee joints were measured with a tape measure and the expression of HSP70 was detected by means of SP immunohistochemical method. Results： Compared with the normal group,the circumference values of bilateral hind-knee joints were increased（ P〈0. 01） and the expression level of HSP70 in the model group was increased（ P〈0. 05）. In comparison with the model group,the circumference values of the bilateral hind-knee joints were decreased significantly and the expression levels of HSP70 after the moxibustion treatment were increased remarkably（ P〈0. 01）,and the effects of the ginger-partitioned moxibustion group were superior to those of other two groups（ P〈0. 05）. Conclusion： Different types of moxibustion treatment may all have an obvious anti-inflammatory effect,and may increase the expression levels of HSP 70 in the rabbits＇ synovial cells. And the treatment of ginger-partitioned moxibustion has a better therapeutic effect probably.
