摘要
目的探讨腺病毒介导白细胞介素-24基因对瘢痕疙瘩成纤维细胞中纤溶酶原激活物抑制剂-1和Ⅰ、Ⅲ型胶原蛋白mRNA及蛋白水平表达的影响。方法构建白细胞介素-24腺病毒载体,感染瘢痕疙瘩成纤维细胞,并设置阴性对照组和空白对照组。应用实时定量PCR检测纤溶酶原激活物抑制剂-1及Ⅰ、Ⅲ胶原蛋白mRNA表达的变化,WesternBlot检测纤溶酶原激活物抑制剂-1及Ⅰ、Ⅲ型胶原蛋白水平表达的变化。结果白细胞介素-24腺病毒载体感染瘢痕疙瘩成纤维细胞后,与阴性对照组相比,感染组纤溶酶原激活物抑制剂-1和Ⅰ、Ⅲ型胶原蛋白mRNA及蛋白表达量降低(P〈0.05)。空白对照组与阴性对照组差异无统计学意义(P〉0.05)。结论白细胞介素-24腺病毒载体可能通过调节纤溶酶原激活物抑制剂.1及Ⅰ、Ⅲ型胶原蛋白mRNA及蛋白的表达,减少瘢痕疙瘩成纤维细胞胶原等细胞外基质的沉积。
Objective To investigate the effect of IL-24 gene on expression on mRNA and protein level of Plasminogen activator inhibitor-1 ( PAI-1 ) , type I collagen and type 111 collagen of human keloid fibrobl asts (KFs). Methods Interleukin 24 (IL-24) gene was cloned into adenovirus vector, then the adenovirus particles expressing IL-24 were infected into keloid fibroblasts (KF cells ) , KF-IL-24; KF-NC and KF-CON were used as the control groups. Real time PCR and Western blot were performed to examine the expression of IL-24 in adenovirus infected cells. Results Compared with KF-CON group and KF-NC group, the expression levels of IL- 24 mRNA and protein were both significantly upregulated in KF-IL-24 group after KF cells were infected ( P 〉 0.05 ). The difference between the KF-CON group and KF-NC group was not statistically significant ( P 〉 0.05 ). Conclusion Adenovirus-mediated IL-24 gene may inhibit deposition of extracellular matrix of KF through con- trolling the expressions of PAI-1, type Ⅰ collagen and type Ⅲ collagen mRNA and protein.
