摘要 通过单因素试验和正交试验对冬虫夏草产分生孢子培养基进行优化,同时探讨培养条件(温度、光照和p H)对分生孢子产量的影响,确定冬虫夏草菌产分生孢子的最佳条件。结果显示:最佳的冬虫夏草产孢子培养基组成为:葡萄糖30 g,酵母粉10 g,Ca Cl2·2H2O 0.3 g,Mg SO4·7H2O 0.5 g,KH2PO41.0 g,Na Cl 1.0 g,Fe SO4·7H2O 0.05 g,Zn Cl20.09 g,组氨酸0.3 g,赖氨酸0.3 g,纯牛奶200 m L,超纯水补充至1 000 m L。最佳培养条件为:14℃、p H5.80、完全黑暗的环境下,培养40 d。在最佳培养基和最佳培养条件下,冬虫夏草单菌落分生孢子产量达到2.98×105个,优化后孢子产量是基础培养基孢子产量的3.6倍,明显提高了分生孢子的产量。 Using the single factor and the orthogonal test, the optimal culture medium for the conidia of Ophiocordyceps sinensis were studied. The effect of temperature,p H and light on the conidia production were also studied. The best culture medium recipe was: glucose 30 g, yeast extract 10 g,Ca Cl2·2H2O 0.3 g, Mg SO4·7H2O 0.5 g,KH2PO41.0 g, Na Cl 1.0 g,Fe SO4·7H2O 0.05 g, Zn Cl20.09 g,histidine 0.3 g lysine 0.3 g,pure milk200 m L, the ultrapure water added to 1 000 m L. The maximum conidia production was produced under the optimal conditions: temperature 14 ℃, p H 5.80 and complete darkness for 40 d. Under the conditions, the maximum conidia production was 2.98×105. After optimization, the conidia production was 3.6 times more than that of the basal culture medium. The conidia production was significantly increased after optimization, which might be beneficial to workload in scale up fermentation later.
