摘要 目的通过检测组织蛋白酶G(Cathepsin G)在人正常椎间盘和退变椎间盘的表达,探讨其与椎间盘退变的相关性。方法实验组67例,椎间盘组织来源于腰椎间盘突出症的患者;对照组62例,椎间盘组织来源于因骨折行前路手术获取的椎间盘。采用HE染色观察椎间盘的退变特征、用免疫组化S-P法检测Cathepsin G蛋白表达,及RT-PCR检测Cathepsin G基因mRNA转录水平。结果在退变椎间盘中,HE染色可见纤维环板层结构非常紊乱,并呈黏液样退变;软骨细胞巢状增生,髓核细胞细胞质成空泡状,周围毛细血管的侵入,炎性细胞的浸润等。免疫组化显示组织蛋白酶G在实验组(89.12±3.41)%明显高于对照组的(11.05±4.59)%,差异有统计学意义(P<0.05);在Cathepsin G基因mRNA转录水平上,实验组为1.463±0.141,对照组为0.674±0.159,差异有统计学意义(P<0.05)。结论 Cathepsin G参与了椎间盘退变的病理改变,与椎间盘退变存在一定相关性。 Objective To explore the correlation between the expression of cathepsin G and lumbar intervertebral disc in humans. Methods The tissue of intervertebral disc lumbar in experimental group were from lumbar disc protrusion disease and the control group were from thoracic- lumber broken. The characteristics of disc degeneration were detected by HE staining,the expression of Cathepsin G protein were detected by immunohistochemistry SP,and the Cathepsin G gene for mRNA transcription level were detected by RT- PCR. Results HE staining showed signs of tissue degeneration in degeneration between intraday. Immunohistochemistry showed that Cathepsin G in the experimental group( 89. 12 % ± 3. 41%) was significantly higher than control group( 11. 05% ± 4. 59%)( P〈0. 05). Compared with the control group,the Cathepsin G gene mRNA transcription level was higher in the experimental group( P〈0. 05). These results were statistically significant. Conclusion Cathepsin G is involved in the pathological changes in disc degeneration.
